Liposomal Delivery Enhances Immune Activation by STING Agonists for Cancer Immunotherapy

Abstract: Overcoming the immunosuppressive tumor microenvironment (TME) is critical to realizing the potential of cancer immunotherapy strategies. Agonists of stimulator of interferon genes (STING), a cytosolic immune adaptor protein, have been shown to induce potent anti-tumor activity when delivered into the TME. However, the anionic properties of STING agonists make them poorly membrane permeable, and limit their ability to engage STING in the cytosol of responding cells. In this study, cationic liposomes with varyin… Show more

“…This is consistent with previous studies that attempted to encapsulate other hydrophilic cargo within PLGA MPs using similar emulsion methods [63, 64]. Compared to other reported CDN delivery vehicles (polymeric hydrogel particles (47%) [42] or liposomes (2 to 35%) [41, 4345]), electrospray was able to achieve a significantly superior CDN EE (90%). This multi-fold increase in EE has promising manufacturing implications for using this electrospray platform to formulate hydrophilic, small molecule CDNs.…”

Exaggerated anticipatory anxiety is common in social anxiety disorder (SAD). Neuroimaging studies have revealed altered neural activity in response to social stimuli in SAD, but fewer studies have examined neural activity during anticipation of feared social stimuli in SAD. The current study examined the time course and magnitude of activity in threat processing brain regions during speech anticipation in socially anxious individuals and healthy controls (HC). Method Participants (SAD n = 58; HC n = 16) underwent functional magnetic resonance imaging (fMRI) during which they completed a 90s control anticipation task and 90s speech anticipation task.

“…cGAMP, selected as an immune response model agonist, and 5′-GMP were then encapsulated into liposomes via thin film rehydration and membrane extrusion to form sub-200 nm sized Lipo-NPs. The encapsulation efficiency of cGAMP was 22.6%, consistent with previous reports . To fabricate liposomes with different elasticities, three identical liposome suspensions were treated with distinct concentrations of Zn(NO 3 ) 2 , cross-linking the cGAMP and 5′-GMP encapsulated cores.…”
