Late histological aspects of spleen autologous transplantation in rats

Purpose:To evaluate macro and microscopically the late evolution of autotransplants of fragments of spleen in the greater omentum, mesenterium and peritoneal cavity, after 24 weeks of observation. Methods: Fifty two Wistar rats were used, males and adults, submitted to total splenectomy and divided in four groups. The group I -seventeen animals with implant of spleen fragment in the peritoneal cavity; group II -eighteen animals with implant in the omentum and group III -seventeen animals with implant fixed in mesenterium root. The group control (group IV) was formed by eight animals chosen aleatorily among the three groups. It was analyzed macro and microscopically the evolution of the implant, and in the histological study qualitative and quantitative criteria were adopted, with the counting of no cellular e cellular elements. Results: It was observed adherences to the adjacent tissues and vascularization in all of the fragments transplanted. The group I presented white pulp and preserved vascularization. In the group II were observed white pulp with follicular formations and lymphoid tissue preserved, and the red pulp in cordon aspect and hemorrhagic. In the group III were observed with depletion of white and red pulp, while others evidenced better preservation of the pulps. The counting of lymphocytes revealed significant difference between the groups I and IV and the group III and IV (p < 0.05). The counting of active macrophages revealed significant difference between the groups II and III (p < 0.05) and similarity between II and IV (p > 0.05). The other elements: active macrophages phagocyting hemosiderine, plasmocytes, fibroblasts, fibrocytes, giant cells, monocytes, interstitial spaces and fibers of collagen, did not show significant difference among the groups. Conclusions: The splenic autotransplantation is feasible, being the better place the greater omentum. This research demonstrated through qualitative and quantitative histological analysis that the splenic tissue autotransplanted in the omentum of Wistar rats preserves its function of defense of the organisms. Epidemiological studies show a higher prevalence of obesity in children from smoking mothers and smoking may affect human thyroid function. To evaluate the mechanism of smoking as an imprinting factor for these dysfunctions, we evaluated the programing effects of maternal nicotine (NIC) exposure during lactation. Two days after birth, osmotic minipumps were implanted in lactating rats, divided into: NIC (6 mg/kg per day s.c.) for 14 days; Control -saline. All the significant data were P!0 . 05 or less. Body weight was increased from 165 days old onwards in NIC offspring. Both during exposure (at 15 days old) and in adulthood (180 days old), NIC group showed higher total fat (27 and 33%). In addition, NIC offspring presented increased visceral fat and total body protein. Lipid profile was not changed in adulthood. Leptinemia was higher at 15 and 180 days old (36 and 113%), with no changes in food intake. Concerning the thyroid status, the 15-days-old NIC offspring showed lower serum-free tri-iodothyronine (FT 3 ) and thyroxine (FT 4 ) with higher TSH. The 180-days-old NIC offspring exhibited lower TSH, FT 3 , and FT 4 ). In both periods, liver type 1 deiodinase was lower (26 and 55%). We evidenced that NIC imprints a neonatal thyroid dysfunction and programs for a higher adiposity, hyperleptinemia, and secondary hypothyroidism in adulthood. Our study identifies lactation as a critical period to NIC programing for obesity, with hypothyroidism being a possible contributing factor.

The procedures in groups 2, 3 and 4 were made simultaneously, and splenectomized animals, groups 2 and 3 were donors, respectively, for the animals of groups 3 and 4. In group 4 the spleen was preserved and the animals received implants from the spleens of rats from group 3. The regeneration of splenic tissue was evaluated by macroscopic and microscopic analyzes of the grafts and own spleens, as well as with measurements of VLDL, HDL, LDL, total cholesterol and triglycerides. Results Results ResultsResults Results: after 120 days, Group 4 showed levels of total cholesterol and LDL lower than the other groups. Group 1 had higher levels of lipids. ResumoNa presente pesquisa foram utilizados 30 espécimes de coelhos adultos Nova Zelândia, 13 machos e 17 fêmeas. Após fixação com solução de formaldeido a 10% e preenchimento do sistema arterial com solução de látex corado procedeu-se a dissecção para a observação da disposição das artérias lienais. A análise estatística foi realizada através teste do X 2 (Qui-quadrado), ao nível de significância de 5%, para testar se a hipótese de nulidade é verdadeira para o número de ramificações principais da artéria lienal. As relações do comprimento da artéria lienal e do baço em função do comprimento rostro-sacral foram calculadas através do coeficiente de correlação "r", que pode variar entre -1 e +1, isto é, -1 ≤ r ≤ +1. A média do comprimento da artéria lienal foi de 2,166 cm. Em todos os coelhos dissecados (100%) a artéria lienal se originou diretamente da artéria celíaca como uma artéria única, emitindo diferentes ramos para o baço: um ramo em sete (23,3%) animais, dois ramos em sete (23,3%) animais, três ramos em 10 (33,3%) animais, quatro ramos em quatro (13,3%) animais e cinco ramos em dois (6,7%) animais. Do baço a artéria lienal emitiu diferentes números de ramos para a grande curvatura do estomago: um ramo em sete (23,3%) animais, dois em quatro (13,3%) animais, três em cinco (16,7%) animais, quatro em três (10%), e cinco em apenas um (3,3%) animal. Foi observada correlação positiva entre o comprimento do baço e da artéria lienal em função do comprimento rostro-sacral. O número de ramificações da artéria lienal independe do sexo do animal. After fixed with 10% formaldehyde solution and performing the injections with latex solution stained with pigment, the animals were dissected to identify the disposition of the lienal arteries. The statistical analysis was made by the Qui-square (X 2 ) test, with a 5% level of significance, to test if the nullity hypothesis is true for the of lienal artery main ramifications. The relations between the lienal artery and spleen length with rostrum-sacral length were calculated by the correlation coefficient "r" varying between -1 and +1, that is, -1 ≤ r ≤+1.
