Lipid metabolites as metabolic messengers in inter-organ communication

Abstract: Metabolic homeostasis is achieved through coordinated regulation across several tissues. Studies using mouse genetic models have shown that perturbation of specific pathways of lipid metabolism in metabolically active tissues impacts systemic metabolic homeostasis. The use of metabolomic technologies combined with genetic models has helped identify several potential lipid mediators that serve as metabolic messengers to communicate energy status and modulate substrate utilization among tissues. When provided ex… Show more

“…Lipid signals influence fat synthesis, storage, or catabolism, creating intricate metabolic networks that allow organisms to respond to nutrient availability, energy demands, or otherwise adapt to changing environments [1]. Storage lipids (triglycerides) and circulating lipid-protein complexes (lipoprotein particles) have previously been linked to diseases of aging [2], and obesity limits longevity [3].…”

“…Interestingly, feeding worms OEA is sufficient to activate these transcriptional regulators and extend lifespan. In mammals, HNF4 and PPARs are key regulators of lipid metabolism, inflammation, and cell death [1,10]. In mice, OEA is induced by feeding, regulates satiety, and activates PPARα [1,9].…”

“…The defenses activated by "noxious" temperature exposure are often different from those activated by "innocuous" changes ( 65 ), and these sensors and reactions may have different impacts on dWAT. When the thermal defense circuit is activated, lipids are continuously mobilized from WAT and BAT, and these circulating lipids are therefore candidates as functional modifi ers of various tissues in cold-exposed mammals, including dWAT, lung, or liver ( 66 ).…”
