Lipid Metabolism and Ferroptosis

Abstract: Ferroptosis is a type of iron-dependent regulated necrosis induced by lipid peroxidation that occurs in cellular membranes. Among the various lipids, polyunsaturated fatty acids (PUFAs) associated with several phospholipids, such as phosphatidylethanolamine (PE) and phosphatidylcholine (PC), are responsible for ferroptosis-inducing lipid peroxidation. Since the de novo synthesis of PUFAs is strongly restricted in mammals, cells take up essential fatty acids from the blood and lymph to produce a variety of PUFA… Show more

“…Then, excessive iron might overpass the cells’ capabilities to maintain iron homeostasis. Our results are consistent with reports about cell death caused by iron treatment at concentrations similar to what we used in our experiments ( Baba et al., 2018 ; Chen et al., 2020 ; Lee et al., 2021 ).…”

“…Furthermore, genes that regulate the production of ROS and lipid peroxides are also identified as biomarker for ferroptosis, such as LOX, NADPH oxidase 4 (NOX4), and ZFP36. The LOX family of enzymes is iron-dependent, the inhibition of which decreases accumulation of lipid peroxides and subsequently blocks ferroptosis [67]. Increased NOX4 impairs mitochondrial function, induces lipid peroxidation, and promotes ferroptosis [68].…”

“…SFA-enriched membranes affect the uptake of certain chemotherapeutics such as doxorubicin and promote the resistance to ionizing radiation ( 35 , 36 ). Since SFAs are more resistant to lipid peroxidation, metastatic PCa cells with SFA-enriched membranes would be most likely less susceptible to oxidative stress-induced ferroptosis, an iron-dependent form of cell death induced by reactive oxygen species (ROS)-mediated lipid peroxidation ( 37 ). Targeting de novo lipogenesis and the Lands cycle has recently been shown to induce ferroptosis in KRAS-mutant lung cancer ( 38 ) and we anticipate similar results in mCRPC.…”
