Abstract: Adult stem cells constitute an important reservoir of self-renewing progenitor cells and are crucial for maintaining tissue and organ homeostasis. The capacity of stem cells to self-renew or differentiate can be attributed to distinct metabolic states, and it is now becoming apparent that metabolism plays instructive roles in stem cell fate decisions. Lipids are an extremely vast class of biomolecules, with essential roles in energy homeostasis, membrane structure and signaling. Imbalances in lipid homeostasis… Show more

“…At 42 dpa, most enriched pathways are related to cell energetic metabolism, as the catabolism of branched-chain fatty acids (BCFA) and branched-chain amino acids (BCAA), suggesting an increased mitochondrial activity ( Figure 11A) (Neinast et al, 2019;Clémot et al, 2020). Active lipolysis of phospholipids and triacylglycerols, reflected in the low abundance of several BCFA species included lysoPC and lysoPE, explain the promotion of ketone body metabolism and the Krebs cycle (TCA) (Grabacka et al, 2016).…”

“…5 A,B, Tables S3 and S4). Increased lipid remodeling and cholesterol biosynthesis has been implicated in enhanced stem cell maintenance, proliferation, and tumorigenesis both in vivo and in organoids 35 , 36 . In our studies, lipid biosynthesis upregulation likely reflects the increased stem cell composition of organoids relative to stromal rich PDXs.…”

“…Lipid metabolism plays a pivotal role in stem cells physiopathology [ 108 , 109 , 110 ]. However, at the moment the number of studies about the lipidome of MSCs is limited, and mainly focused on variations in lipid composition during stem cell proliferation and differentiation [ 111 , 112 , 113 , 114 , 115 , 116 , 117 , 118 , 119 , 120 , 121 , 122 , 123 , 124 , 125 , 126 , 127 , 128 , 129 , 130 ] ( Figure 2 ).…”

“…More and more reports have been released regarding the lipid profile of MSCs recently, and the role of lipoproteins in altering MSC functions and properties, including their proliferation capacity, immune-modulatory functions, immunomodulatory regulation, and the differentiation potential of multiple lineages [ 47 , 48 ], although most of these investigations have focused on the effect of lipids in regulating MSCs’ adipogenic differentiation capability. However, uncertainty persists regarding the relevance of MSC differentiation through cardiomyogenesis and vasculargenesis.…”

“…Lipidomics is one of the most recent, rapidly developing and promising approaches [ 13 ]. Lipidomics studies the state of the lipid molecular phenotype, reflecting the functional “landscape” of lipid activity in cells and tissues [ 14 ]. For this reason, clinical lipidomics offers the possibility of elucidating the strategic roles of lipids in disease and the immune system [ 15 ], identifying biomarkers and developing new therapeutics [ 16 ].…”
