Leukotriene B4 receptor 2 gene polymorphism (rs1950504, Asp196Gly) leads to enhanced cell motility under low-dose ligand stimulation

Abstract: Recently, single-nucleotide polymorphisms (SNPs) in G-protein-coupled receptors (GPCRs) have been suggested to contribute to physiopathology and therapeutic effects. Leukotriene B4 receptor 2 (BLT2), a member of the GPCR family, plays a critical role in the pathogenesis of several inflammatory diseases, including cancer and asthma. However, no studies on BLT2 SNP effects have been reported to date. In this study, we demonstrate that the BLT2 SNP (rs1950504, Asp196Gly), a Gly-196 variant of BLT2 (BLT2 D196G), c… Show more

“…One of the major activities of LTB 4 is the recruitment of leukocytes, including neutrophils 18 . LTB 4 exerts its biological functions through two G protein-coupled receptors (GPCRs), BLT1 and BLT2 19 . BLT1, a high-affinity receptor for LTB 4 , is mainly expressed in inflammatory cells, such as leukocytes.…”

“…KLK15 is a peptidase and involved in the regulation of cancer cell growth, migration of cancer cells and immune regulation [65]. The LTB4R2 gene is a pro-inflammatory lipid mediator that is only overexpressed under stress-induced inflammatory conditions [66] and plays an important role in inflammatory diseases such as cancer and asthma [67]. Although the receptor 2A was upregulated, the LTB4R gene was strongly downregulated in the IST group.…”

“…This proinflammatory molecule has two receptors, BLT1 and BLT2 ( 14 ). These receptors are members of the G protein-coupled receptor (GPCR) protein and are expressed on cell surface ( 14 - 16 ). BLT1, a high-affinity receptor of LTB 4 , is exclusively expressed on the surface of leukocytes.…”
