BackgroundWith the aim of studying Leptospira spp. infection in sheep herds, blood samples and respective kidney and liver fragments were collected from 100 animals from twenty different properties during slaughter at a meat company in the Sorocaba region, São Paulo state, southeast Brazil. The microscopic agglutination test (MAT) was performed with 29 strains of Leptospira spp. To identify the agent in the liver and kidney, 100 samples of each tissue were submitted to culture in Fletcher medium and analyzed by the polymerase chain reaction (PCR) for Leptospira spp.ResultsMAT detected 23 samples serologically positive for one or more Leptospira spp. serovars and significantly more for Autumnalis. Eight (4%) samples were positive in culture (four kidneys and four livers), corresponding to five animals with positive serology (one animal simultaneously positive for both kidney and liver) and two negatives. PCR detected Leptospira spp. in 14 samples (seven kidneys and seven livers) corresponding to 12 positive animals (two animals simultaneously positive for kidney and liver), of which ten were serologically positive and two negative.ConclusionsPCR was faster, more practical and more sensitive than culture for detecting leptospires. The results reinforce the importance of sheep in the epidemiological context of leptospirosis.

In order to assess the diffusion of brucellosis by Brucella ovis and leptospirosis in sheep flocks in Tuscany, 410 blood samples were collected from males of 76 breeding farms from January to December 2015. All sera resulted negative for B. ovis. A percentage of 17.80% sera was found positive for Leptospira spp. Among all breeding farms, 34.21% resulted positive. The most represented serovars were Pomona (6.34%), Hardjo (4.14%), Grippotyphosa (3.17%) and Bratislava (1.70%). The highest antibody titers were detected for serovar Hardjo (1:25600) and serovars Pomona and Grippotyphosa (1:1600). These results confirm the role of sheep as maintenance host for serovar Hardjo and highlight the spreading of serovar Grippotyphosa in the study area. Constant field investigation, especially on farm animals, could be useful to determine trends and diffusion of some occupational re-emerging diseases, such as leptospirosis.

O objetivo desse estudo foi determinar a ocorrência e os fatores associados à soropositividade para Leptospira spp. em ovinos do estado de Sergipe, Brasil, a partir de 1.200 amostras coletadas de sessenta propriedades das suas três mesorregiões entre os anos de 2011 e 2012. Foi observada, pela reação de soroaglutinação microscópica, 37,7% (452/1.200; IC: 0.357-0.396) de ovinos soropositivos com 93,3% (56) de propriedades com pelo menos um animal positivo. Os sorogrupos reagentes predominantes foram: Sejroe (32,1%), Icterohaemorrhagiae (23,5%) e Grippotyphosa (22,4%), com presença também de reações para Australis (7,1%), Pomona (3,8%), Autumnalis (3,5%), Panama (3,3%), Ballum (1,8%), Andamana (1,3%), Hebdomadis (0,9%) e Seranga (0,4%). As titulações variaram de 100 a 3.200, sendo que 77,2% delas ≤ 400. Os fatores associados à presença de ovinos soropositivos a Leptospira spp. foram: criações de subsistência (p=0,000, OR=3,08, IC95%=1,99-4,78) e com finalidade de produção de matrizes e reprodutores (p=0,000, OR=2,19, IC95%=1,46-3,28). As infecções por Leptospira spp. estão ostensivamente presentes nos rebanhos ovinos do estado de Sergipe, sendo necessária a adoção de práticas de manejo adequadas, direcionadas ao controle de roedores e criação consorciada com bovinos que podem levar a contaminação dos alimentos e pastagens.
