Studies of patients with major depressive disorder (MDD) and schizophrenia (SCH) have revealed reduced hippocampal volumes, but findings have been inconsistent due to sample and measurement differences. The current study sought to measure this structure in a large sample of MDD, SCH, and healthy subjects, using a strict measurement protocol, to elucidate morphological-specific volumetric differences. Patients with treatment-resistant MDD (N = 182) and treatment-resistant SCH with auditory-verbal hallucinations (N = 52), and healthy controls (N = 76) underwent psychiatric assessments and brain MRI. The findings indicate that (1) MDD and SCH patients have reduced total hippocampal volume which was marked in the tails (more so in patients with MDD), (2) region of interest estimation protocols and sample characteristics may help explain volumetric differences between previous SCH studies.

The study's aim was to assess a broad range of declarative and nondeclarative memory functions in schizophrenia to identify areas of impairment versus relative preservation. Participants included 40 schizophrenia outpatients and 30 demographically comparable community residents. All participants were administered a battery assessing declarative memory (verbal learning, working memory, semantic memory, remote memory, verbal retention) and nondeclarative memory (procedural learning, priming). To control for order effects, the battery was divided into three parts of approximately equal length with order of administration counterbalanced across study participants. The results showed persons with schizophrenia to be significantly impaired relative to community residents in verbal learning, working memory, semantic memory, remote memory, and priming. In contrast, the two groups were comparable in verbal retention and procedural learning. In the schizophrenia group, priming ability best discriminated past year's vocational status. In sum, the findings indicate a specific pattern of impairment and preservation of memory functioning in schizophrenia. Skill (procedural) learning and retention of learned, declarative verbal information across a delay appear intact, while all other areas measured appear impaired.

Structural magnetic resonance imaging and postmortem studies showed volume loss in the hippocampus in schizophrenia. The noted tissue reduction in the posterior section suggests that some cellular subfractions within this structure might be reduced in schizophrenia. To address this, we investigated numbers and densities of neurons, oligodendrocytes and astrocytes in the posterior hippocampal subregions in postmortem brains from ten patients with schizophrenia and ten matched controls using design-based stereology performed on Nissl-stained sections. Compared to the controls, the patients with schizophrenia showed a signiWcant decrease in the mean number of oligodendrocytes in the left and right CA4. This is the Wrst Wnding of reduced numbers of oligodendrocytes in CA4 of the posterior part of the hippocampus in schizophrenia. Our results are in line with earlier Wndings in the literature concerning decreased numbers of oligodendrocytes in the prefrontal cortex in schizophrenia. Our results may indicate disturbed connectivity of the CA4 of the posterior part of the hippocampus in schizophrenia and, thus, contribute to the growing number of studies showing the involvement of posterior hippocampal pathology in the pathophysiology of schizophrenia.
