Knockdown of SNORA47 Inhibits the Tumorigenesis of NSCLC via Mediation of PI3K/Akt Signaling Pathway

Abstract: BackgroundNon-small cell lung cancer (NSCLC) is a frequently diagnosed aggressive cancer all over the world. Small nucleolar RNAs (snoRNAs) are a group of non-coding mediatory RNAs. A previous report indicated that small nucleolar RNA 47 (SNORA47) is upregulated in NSCLC. However, the role of SNORA47 in NSCLC is unclear.Material and MethodsCell proliferation was measured by immunofluorescence staining. Cell apoptosis and cycle of NSCLC were tested by flow cytometry and the protein expressions were investigated… Show more

“…Emerging evidence suggests snoRNA's involvement in metabolism, diabetes, cancer, and heart development (20,21,23,34). Specifically, dysregulation of SNORA47, SNORA74A, and SNORNA19 was previously described to be differentially expressed in different cancers (35)(36)(37). Further, SNORA74A was differentially expressed in the myocardium derived from infants with tetralogy of Fallot (38).…”

“…Since SnoRNA is involved in the development of cancer in so many ways, many researchers are eager to explore its potential as a target for cancer treatment. As mentioned above, knocking down SNORA42 can prevent NSCLC to some extent, and may be an effective therapeutic target [40].…”

“…Some snoRNAs are closely related to these two signaling pathways. Knocking down SNORA47 can signi cantly inhibit the occurrence of NSCLC by inhibiting the PI3K-AKT signaling pathway [40]. SnoRNA ACA11 can activate PI3K-AKT pathway to promote cell growth, migration and invasion, and participate in the occurrence of HCC [41].…”

“…However, most of the in vitro studies were performed using primary tumours with a bone tropism, suggesting a role of snoRNAs in metastatic progression in bone. Indeed, experimental modulation of snoRNAs has been shown to regulate migration/invasion in different cancer cell lines derived from prostate (SNORA42, SNORA55) [88,89], breast (SNORA7B, SNORA71A, SNORD50A/B) [90][91][92], lung (SNORA42, SNORA47, SNORA71A, SNORD38, SNORD78) [86,[93][94][95][96], and ovarian cancer (SNORA70E, SNORA72, SNORD89) [97][98][99]. In addition, some of these snoRNAs have also been shown to modulate stemness capabilities in these different cancer types having a bone metastatic tropism (SNORD78, SNORA42, SNORD89, SNORA72, SNORA71A).…”
