Platelet-rich plasma (PRP) is expected to be an alternative therapy in wound healing by accelerating the inflammatory process and increasing the healing factors so that the healing process or inflammation is faster. The purpose of this study was to examine the effect of calcium chloride (CaCl2)-activated PRP on the viability, migration, and cytokine levels of interleukin-6 (IL-6) and vascular endothelial growth factor (VEGF) of dermal fibroblast cells in lipopolysaccharide (LPS)-induced inflammatory conditions, as a model in wound healing in vitro. Fibroblast cells were grown in DMEM medium induced with LPS, and then CaCl2-activated PRP treatment was added. Measure fibroblast cell viability using CCK-8 kit (cell counting kit-8) was evaluated using a microplate reader, and the cell migration was evaluated using scratch-assay and TScratch software. Expression of IL-6 and VEGF using ELISA kit. All data were analyzed using software SPSS version 26 by performing a one-way analysis of variance (ANOVA), Kruskal Wills, and Mann-Whitney tests. The results showed that PRP significantly increased fibroblast cell viability in the 10% PRP treatment group. This study shows that PRP does not reduce IL-6 cytokine levels but can increase VEGF growth factor in fibroblast cell cultures. PRP increased cell migration so that the healing process was faster. In conclusion, the CaCl2-activated PRP on LPS-induced fibroblast cells can increase viability and accelerate cell migration; it can’t decrease IL-6 but can increase VEGF expression. PRP is expected to be an alternative therapy in wound healing.

AbstrakBerbagai penelitian ilmiah telah membuktikan bahwa kolagen merupakan biomaterial ideal dalam aktivitas penyembuhan luka. Senyawa bioaktif pada madu dan Aloe vera dapat membantu meningkatkan pembentukan kolagen. Penelitian ini dilakukan untuk membandingkan pengaruh aplikasi topikal madu, gel Aloe vera, dan Moist Exposed Burn Ointment (MEBO) terhadap kepadatan kolagen pada proses penyembuhan luka bakar derajat dua pada tikus. Luka bakar dibuat pada 28 ekor tikus dan dibagi menjadi empat kelompok yang dipilih secara acak serta dilakukan aplikasi topikal harian dengan NaCl, madu, gel Aloe vera, dan MEBO secara berurutan. Biopsi kulit dilakukan pada hari ke-7, kemudian dilakukan pembuatan sediaan histopatologi kulit dan dihitung kepadatan kolagennya. Uji One Way ANOVA menunjukkan kepadatan kolagen berbeda secara signifikan antar masing-masing kelompok (p= 0,009). Uji Post Hoc Bonferroni menunjukkan terdapat perbedaan yang signifikan antara kelompok NaCl dengan madu (p= 0,024) dan NaCl dengan MEBO (p= 0,024). Penelitian ini menunjukkan bahwa aplikasi madu dan MEBO secara topikal pada luka bakar derajat dua dapat meningkatkan pembentukan kolagen, sehingga dapat mempercepat proses penyembuhan luka. Madu, Aloe vera, dan MEBO dapat dijadikan sebagai terapi alternatif dalam penyembuhan luka bakar. AbstractVarious scientific studies have proven that collagen is an ideal biomaterial in wound healing activities. The bioactive compounds in honey and Aloe vera can help increase collagen formation. This study was conducted to compare the effect of topical application of honey, Aloe vera gel, and MEBO (Moist Exposed Burn Ointment) on collagen density in the healing process of second-degree burns in rats. Burns were made on 28 rats which were further randomly divided into four groups to receive daily topical application of NaCl, Aloe vera, honey, and MEBO respectively. Skin biopsy was carried out on the seventh day, then histopathological preparation of the skin was made and collagen density was calculated. Result of One Way ANOVA test showed that collagen density differed significantly between groups (p= 0.009). The Post Hoc Bonferroni test resulted in significant difference between NaCl with honey group (p= 0.024) and NaCl with MEBO (p= 0.024). This study found that topical application of honey and MEBO to second-degree burns could increase collagen formation, thus accelerating wound healing process. Honey, Aloe vera, and MEBO can be used as alternative therapies for healing burns.

Background and Aim: The virulence and antifungal resistance of Candida albicans are recently known for their ability to form biofilm. This research aimed to construct an in vivo model of C. albicans biofilm in Wistar rats' intestinal mucosa and study their mobilization while in a planktonic and biofilm formation. In this study, there was one treatment group that was treated with three antibiotics, immunosuppressants, and C. albicans. Materials and Methods: This study was divided into control and treatment groups. The data sampling was conducted after C. albicans inoculation. The C. albicans biofilm formation stage was monitored with colony-forming units method calculation every week post-inoculation and then observed by the confocal laser scanning microscope. Results: The planktonic C. albicans overgrowth occurred up to 14 days after inoculation. The formation and maturation of C. albicans biofilm in the intestinal mucosa started in the 28th and 35th-day post-inoculation, respectively. The density of planktonic C. albicans in the stool was dramatically decreased on the 35th day. Before the biofilm formation, the planktonic Candida was carried away by food scraps to be released as a stool. However, there were minuscule or no planktonic Candida observed in the stool during and after biofilm formation. Instead, they were attached to the caecum's mucosa as a biofilm. Conclusion: We have proved that the planktonic C. albicans with its mobile nature were carried into the stool along with the rest of the feed, as we observed a lot of C. albicans cells found in the stool. Meanwhile, on day 28 after administration of antibiotics and immunosuppressants, no C. albicans was found in the stool samples, and at the same time, we observed C. albicans cells and their matrix attached to the intestinal mucosa as a biofilm.
