Ivy/neurogliaform interneurons coordinate activity in the neurogenic niche

Abstract: Depolarization by the neurotransmitter GABA regulates adult neurogenesis. Here we show that interneurons of the neurogliaform cell family are a primary source of GABA for newborn neurons in mouse dentate gyrus. GABAergic depolarization occurs in concert with reduced synaptic inhibition of mature neurons, suggesting the local circuitry facilitates coordinated activation of new and pre-existing cells.

“…Ivy cells are the quantitatively most abundant type of GABAergic interneuron in the CA1 area 25 . They provide widespread synaptic and extrasynaptic slow GABAergic input, as well as NPY and nitric oxide to pyramidal cell dendrites and Schaffer collateral/commissural terminals via their exceptionally dense axonal arborizations 25,26 and may mediate the activity-dependent regulation of neurogenesis 43 . The firing patterns identified here, are well suited to control homeostasis in the network.…”

“…The first synaptic inputs onto new granule neurons are GABAergic, and running, which accelerates granule cell maturation, enhances GABA in the dentate gyrus (Markwardt et al, 2011;Snyder et al, 2009b). In fact, the anxiolytic-like effects of running are blocked by GABA receptor antagonists in mice , suggesting that running and anxiolytic drugs may share the same mechanism.…”

“…The order in which these inputs become functionally connected to abGCs has been the subject of considerable study ( (Markwardt et al 2011). GABA is depolarizing at this time (due to immature, high internal chloride concentration), but when exogenously applied inhibits cell firing via shunting (Overstreet-Wadiche et al 2005).…”
