SummaryAn enzyme-linked immunosorbent assay (ELISA) to measure Streptobacillus moniliformis antibodies in mice and rats was developed. Twelve S. moniliformis strains originating from cases of rat-bite fever and Haverhill fever in man and from various rodent species, showed considerable serological relationship.The ELISA appeared specific since antibodies to S. moniliformis were absorbed by autologous and homologous antigen, but not by heterologous bacterial antigens.Acholeplasma laidlaw;; showed partial serological relationship with S. moniliformis.The ELISA was validated using experimental infections in mice and rats. These studies and observations in naturally infected feral rats, confirmed that S. moniliformis is difficult to grow on primary isolation, and that the ELISA for S. moniliformis antibodies revealed more contaminated animals than culture. ABSTRACT. We isolated Streptobacillus moniliformis, the causative agent of rat-bite fever in humans, from the salivary gland of a pet rat postmortem. The isolate was a Gram-negative pleomorphic coccobacillus, which produced acid from glucose and showed enzymatic activities for eight items in the API ZYM system. The results were consistent with those of the reference strain, ATCC 14647 T , except for acid production from dextrin. Partial sequencing of 16S rRNA (1,440 bp) and gyrB genes (514 bp) of the isolate revealed similarities of 100% and 99.8%, respectively, to those of S. moniliformis in GenBank. Therefore, the isolate was identified as S. moniliformis. These results suggested the potential risk of rat-bite fever arising from pet rats in Japan.

Gram‐stain‐negative. Conversion to L ‐phase or transitional‐phase variant may occur spontaneously during cultivation. Capable of growth anaerobically or aerobically. Ferments glucose to produce acid but not gas . Optimum temperature 35–37°C. Catalase and oxidase negative. Indole not produced. Nitrate not reduced to nitrite. Require serum, ascitic fluid, or blood for growth. Isolated from the throat and nasopharynx of wild and laboratory rats. Causes rat‐bite fever in man . DNA G + C content ( mol %): 24–26.
