Isolation of an RNA-Directed RNA Polymerase–Specific cDNA Clone from Tomato

Abstract: A 3600-bp RNA-directed RNA polymerase (RdRP)-specific cDNA comprising an open reading frame (ORF) of 1114 amino acids was isolated from tomato. The putative protein encoded by this ORF does not share homology with any characterized proteins. Antibodies that were raised against synthetic peptides whose sequences have been deduced from the ORF were shown to specifically detect the 127-kD tomato RdRP protein. The immunoresponse to the antibodies correlated with the enzymatic activity profile of the RdRP after chr… Show more

“…RdRPs amplify an siRNA signal by generating double-stranded RNA (dsRNA) from single-stranded RNA 25 , but Drosophila and other insects lack RdRP genes. RdRP is present in some ticks 26 , and similarly, we identified RdRP genes across the chelicerates, frequently in multiple copies (Fig.…”

“…Reaction (1) presumably involves initiation at the 3′ end of aberrant mRNA and processive RNA synthesis, whereas reactions (2) and (3) would require internal recruitment of the polymerase to its RNA targets. Both initiation modes function in vitro [9,24]. The more open conformation of subunit B would allow internal initiation, but conversely, the partial closure of the RNA product groove in the dimer might favour initiation at the 3′ end of an RNA template.…”

“…RDRs enhance the potency of RNAi by amplifying the aberrant RNA population. It is defined by the presence of a conserved RNA-dependent RNA polymerase catalytic domain and is required for initiation and amplification of the silencing signal (Schiebel et al, 1998). Multiple copies of AGO and RDR genes are known to exist in both plants and animals.…”
