Over the past few decades, South American Rivers, including the Paraná River system, have been intensely impacted by dam constructions. To mitigate adverse impacts, fish passages were incorporated into two large dams on the Paraná River (Itaipu and Porto Primavera) to reestablish longitudinal connectivity, which is very important for conserving populations of migratory fish species. The aim of this study was to estimate genetic diversity, population differentiation and gene flow, using microsatellites markers and mitochondrial haplotypes (D-loop), in populations of Prochilodus lineatus, a long distance migratory species commonly found in Paraná River basin drainages. Samples were obtained from six locations in the Upper and Middle Paraná River: Porto Primavera Reservoir, downstream of the Porto Primavera Reservoir, four tributaries of this reservoir (Pardo, Peixe, Verde and Aguapeí Rivers) and in the Iguaçu River, downstream of the Itaipu dam. High genetic diversity was observed in all samples, as expected for species with large population sizes and high migration rates. However, the influence of other factors, such as secondary contacts, fish passages and restocking were also suggested. In general, evidence of spatial population structuring was not found, even though there were two large dams in the study area. This could also be attributed to the brief period that has elapsed since the dams were built, the influence of migratory and reproductive patterns and the possible contributory effects of the fish passages. Furthermore, the absence of population structuring also corroborates existing information that emphasizes the importance of unimpeded tributaries for conserving migratory fish populations in fragmented watersheds.

Neotropical fish correspond to approximately 30% of all fish species worldwide. The diversity of fish species found in Neotropical basins reflects variations in life-history strategies and exhibition of particular morphological, physiological and ecological attributes. These attributes are mainly related to different forms of feeding, life maintenance and reproduction. Today, fish populations are being threatened by anthropogenic actions that are having a visible impact on the natural state of continental aquatic ecosystems. The main causes are overfishing, non-native species introduction, reservoir-dam systems, mining, pollution and deforestation. The biology and population dynamics of the species are still unclear due to lack of research. Genetic tools can be useful resources for the conservation of Neotropical fish species in several ways. Molecular genetic markers are considered powerful tools to identify cryptic and hybrid fish and also allow the evaluation of the genetic variability and structure of populations of Neotropical ichthyofauna. Several analyses of molecular markers have been performed on Neotropical fish, including allozyme analysis, restriction fragment length polymorphisms in regions of DNA (RFLP), randomly amplified polymorphic DNA (AFLP), randomly amplified polymorphic DNA (RAPD), microsatellites, single nucleotide polymorphisms (SNPs) and mitochondrial DNA (mtDNA) markers. ResumenEl Ichthyoelephas longirostris comúnmente conocido como el Pataló es una especie endémica de la cuenca del río Magdalena y del río Ranchería. Posee una fuerte presión pesquera comercial y de consumo. Según el libro rojo de peces dulceacuícolas de Colombia actualmente se encuentra reportada como una especie amenazada debido a las fuertes alteraciones de su hábitat producidas principalmente por contaminación y erosión. Teniendo en cuenta su importancia y la falta de información genética reportada, en este estudio se analizaron seis marcadores microsatélites para determinar el grado de variabilidad y estructura genético-poblacional del Pataló de los ríos La Miel y Ranchería. Para las dos localidades la heterozigosidad esperada varió de 0,6182 a 0,546, la heterozigosidad observada entre 0,6812 a 0,454, la riqueza alélica entre 6,760 y 4,640 y la diversidad génica encontrada estuvo entre 0,631 y 0,540 respectivamente. El índice de endogamia FIS evidenció un exceso de heterocigotos dado por su valor medio negativo. Los análisis realizados indican que existen dos poblaciones diferentes de Pataló con una estructuración genética moderada, las cuales presentan un buen estado genético. Estos resultados son de gran valor por ser el primer estudio de caracterización genética realizado en esta especie y con el cual se podrán tomar medidas de control y manejo especialmente cuando ya se están implementando acciones de repoblamiento y se está trabajando en su reproducción en cautiverio.Palabras claves: Peces de agua dulce, estructura poblacional, conservación, variabilidad genética.
