Is TIMP‐1 a biomarker for periodontal disease? A systematic review and meta‐analysis

Abstract: Objective One of the most important families of proteases associated with periodontal disease is the family of the matrix metalloproteinases (MMPs). Their activity is regulated by tissue inhibitors of metalloproteinases (TIMPs), and an imbalance between MMP activity and regulation by TIMPs has been associated with the progression of periodontal disease. This strong interaction between TIMPs and MMPs might be an indication that TIMPs can be used as a biomarker to monitor periodontal disease progression in oral … Show more

“…TIMP-1 has a more restricted inhibitory range than the other TIMPs, also strongly binds to both active and zymogen of MMP-9 than to MMP-8. An imbalance between the MMP-8, MMP-9, and TIMP-1 subsequently drives the various pathologic disorders (Bostanci et al, 2021;Vandenbroucke & Libert, 2014).…”

“…Furthermore, HNE can proteolytically inactivate TIMP-1 (Jackson et al, 2010). An imbalance between MMP-8 and TIMP-1 can lead to tissue destruction and functional alterations, thereby leading to destructive inflammatory diseases (Bostanci et al, 2021;Lahdentausta et al, 2018). Enzymes among MPO and HNE can also potentiate the destructive MMP cascade (Nizam et al, 2014).…”

“…The imbalance of MMP and TIMP-1 in the early stages of the pathological process indicates the greatest expediency of using MMP inhibitors in the treatment of patients with gingivitis and periodontitis of the initial degree. У свою чергу, активність ММП регулюється специфічними тканинними інгібіторами ТІМР (Tissue Inhibitor of Metalloproteinase), а дисбаланс між ними зв'язують із перебігом остеорезорбтивних процесів у тканинах пародонта [9]. Установлено специфічність дії ТІМР-1 до ММП-8 та 129 22/ Том XXVII / 3 ММП-9 у хворих на пародонтит, проте цей інгібітор здатний пригнічувати й решту ММП [7].…”

“…6 TIMP-1 is known as an inhibitor of matrix metalloproterinase. 7 Saliva contains biomarkers specific to periodontitis and thus qualitative changes in the composition of these biomarkers can be diagnosed. Experimental evidence suggests that the most important pathway involving MMP as active collagenase and gelatinase is found not only in gingival sulcus fluid but also in saliva and biopsy specimens in inflammatory periodontal tissue.…”
