Abstract: Contact sites of endoplasmic reticulum (ER) and mitochondria locally convey calcium signals between the IP 3 receptors (IP3R) and the mitochondrial calcium uniporter, and are central to cell survival. It remains unclear whether IP3Rs also have a structural role in contact formation and whether the different IP3R isoforms have redundant functions. Using an IP3R-deficient cell model rescued with each of the three IP3R isoforms and an array of super-resolution and ultrastructural approaches… Show more

“…The number of ER-mitochondrial contacts depends on the expression of the IP3R Ca 2+ channel [100]. This channel forms a tether with VDAC via the mitochondrial chaperone glucose-regulated protein 75 (GRP75).…”

“…As a major Ca 2+ release channel on ER, the largest intracellular Ca 2+ store in mammalian cells [7], IP 3 Rs often form large complexes with other Ca 2+ handling proteins and Ca 2+ channels [8]. Numerous reports have showed biochemical or co-localization evidence indicating that IP 3 Rs interact with PM Ca 2+ -transporting ATPase (PMCA) [9,10], Sarco/endoplasmic reticulum Ca 2+ ATPase 2 (SERCA2) or SERCA3 [10,11], STIM1 [12], Orai1 channels [13,14], and handling organelles like mitochondria [15] in different types of cells. Thus, IP 3 Rs are key elements of Ca 2+ signalling machinery.…”

“…1C). Transmission of calcium into mitochondria depends on which ITPR isoforms are expressed at the MAM, ( 15,16 ) which varies among cell types and can change in disease states. ( 13,14,17,18 ) Therefore, we examined whether expression of each of the three ITPR isoforms was altered in YF.…”

“…( 12‐14,16 ) Normally this is regulated by ITPR1 in hepatocytes, ( 14 ) and expression of this isoform was not altered by YFV. ITPR2 can preferentially transmit calcium into mitochondria in some cell types, ( 15,40 ) and ITPR2 expression was increased in YFV‐infected hepatocytes, but there is disagreement about whether this isoform localizes to the MAM and does not regulate steatosis in hepatocytes. ( 14,41 ) ITPR3 can preferentially transmit calcium into mitochondria in other cell types ( 15‐17,42 ) .…”
