The aim of this study was to investigate antioxidant activity, total phenolic compounds and flavonoids of propolis from three provinces of Indonesia with two methods of extraction. Stingless bee propolis from Banten, South Kalimantan, and South Sulawesi was prepared through ultrasound-assisted extraction with water and ethanol solvents, separately. Antioxidant activity was measured by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) method. Total phenolic and flavonoid contents were analyzed by Folin-Ciocalteu colorimetric and aluminum chloride (AlCl3) methods, respectively. The result showed that the antioxidant activity expressed as IC50 of Indonesian propolis ranged from 452.52 to 1027.29 mg/L. Ethanol extracts showed higher antioxidant activity than water extracts. Total phenolic and flavonoids content detected in the samples ranged from 10.00 to 28.65 mg/g GAE and 0.76 to 3.39 mg/g QE, respectively. The ethanol extract of propolis from South Kalimantan had the highest total phenolic contents, while that from South Sulawesi possessed the highest total flavonoids.

Bees produce propolis, a mixture of bee saliva and gum produced by leaf buds and stems, which come out through plant's skin collected by bees. 1 The physical properties of raw propolis are hard and wax-like when fresh, but soft and very sticky when warm. It has a characteristic aromatic smell and odor; its range color varies from light to dark brown, red, yellow, or green, depending on its age and source.2 Chemical composition, which bees collect from the resinous plant parts, may influence its biological effect. 3 The composition of propolis depends on diverse vegetation, phytogeographic region, and time of the collection. 4 Results from previous researchers found more than 300 components contained in propolis i.e. aromatic acids; aromatics esters; flavanones; flavones and flavonol; chalcones and dihydrochalcones; terpenoids; acyclic hydrocarbons and esters; alcohols; aliphatic acids (short-chain); aliphatic esters; aliphatic fatty acids (long-chain) and esters; amino acids; aromatic hydrocarbons; acetophenones and other ketones; glycerol derivatives; steroids; sugars and sugar alcohols; and miscellaneous ingredients. The propolis ethanolic extracts (PEE) was dried and studied their antioxidant properties by using FRAP (Ferric Reducing Antioxidant Power) and DPPH radical scavenging assays. Total phenolic compounds were quantified by Folin-Ciocalteu, and total flavonoid contents were also quantitatively determined by the AlCl 3 colorimetric method with a microplate reader. The chemical compounds were identified by an ultraperformance liquid chromatography TOF mass spectrometer (UPLC-TOF-MS) using the MS E mode.

The current study provided a comparison the antioxidant activity, total phenolic and total flavonoid contents of propolis in two different types of honey bees and location. The material used for this research were propolis which was produced by different honey bees, namely Apis mellifera and Trigona sp in different location from Batu and Mojokerto. The method used was descriptive analysis. Findings suggested the maximum and minimum of antioxidant activity were obtained by propolis Trigona sp. from Mojokerto (987.24 µg/g) and propolis Trigona sp. from Batu (166.25 µg/g). The maximum and minimum of total flavonoid content were obtained by propolis Apis mellifera from Mojokerto (1.990 mg/g) and propolis Trigona sp from Mojokerto (1.000 mg/g). The maximum and minimum of total fenolic content were obtained by propolis Apis mellifera from Mojokerto (21.980 mg/g) and propolis Trigona sp from Mojokerto (9.603 mg/g). Propolis Apis mellifera from Mojokerto had a higher total flavonoids and phenolic content and Trigona sp. from Mojokerto had lowest of antioxidant activity, total flavonoids and total phenolics.
