Intracellular Concentrations of Borrelia burgdorferi Cyclic Di-AMP Are Not Changed by Altered Expression of the CdaA Synthase

Abstract: The second messenger nucleotide cyclic diadenylate monophosphate (c-di-AMP) has been identified in several species of Gram positive bacteria and Chlamydia trachomatis. This molecule has been associated with bacterial cell division, cell wall biosynthesis and phosphate metabolism, and with induction of type I interferon responses by host cells. We demonstrate that B. burgdorferi produces a c-di-AMP synthase, which we designated CdaA. Both CdaA and c-di-AMP levels are very low in cultured B. burgdorferi, and no … Show more

“…The levels of c-di-AMP in the DAC overexpression strain were significantly altered, i.e., 1.7-fold higher ( p < 0.05), compared to the WT strain. Whereas this is lower than might be anticipated based on gene expression data, some bacterial strains which overaccumulate DAC mRNA show no upregulation of intracellular c-di-AMP levels suggesting possible negative feedback mechanisms on DAC activity in some cases ( Savage et al, 2015 ). The levels of c-di-AMP in the PDE overexpression Synechocystis strain were on average half as much as the WT strain, although this level was not significantly different.…”

“…There is limited but significant evidence that c-di-AMP is produced by (some) Gram-negative organisms. This has been noted in Chlamydia trachomatis [ 31 ] and Borrelia burgdorferi [ 32 , 33 ].…”

“…The flaB transcript is frequently used as a qRT-PCR reference transcript [ 58 , 69 , 70 ]. The recA mRNA has also been used on occasion as a reference, under the same assumption of invariability [ 16 , 18 ]. However, our RNA-Seq analyses consistently revealed differences in both flaB and recA mRNA levels when comparing early or mid-exponential to stationary phases ( S4 and S5 Tables).…”
