Abstract: The immune-modulating cytokine interleukin-6 (IL-6) is expressed both in adipose tissue and centrally in hypothalamic nuclei that regulate body composition. We investigated the impact of loss of IL-6 on body composition in mice lacking the gene encoding IL-6 (Il6-/- mice) and found that they developed mature-onset obesity that was partly reversed by IL-6 replacement. The obese Il6-/- mice had disturbed carbohydrate and lipid metabolism, increased leptin levels and decreased responsiveness to leptin treatment. … Show more

“…Dosedependent increases in total cholesterol, HDL cholesterol, and triglycerides were observed and were above the normal range in some patients. This finding is concordant with previous reports that administration of recombinant IL-6 decreased serum cholesterol in cancer patients (31,32) and that IL-6-deficient mice showed an increase in triglycerides and very-low-density lipoprotein in the blood in association with suppressed energy expenditure and increased food intake, with no increase in HDL cholesterol (33). However, MRA treatment also increased HDL cholesterol.…”

“…Taken together, these results indicate that β-adrenergic/G protein α subunit/cAMP signalling in adipocytes is an important regulator of whole-body homeostasis. Recently, several inflammatory cytokine-deficient mice, including Il6 −/− [44], Il1r1 −/− [11], Csf2 −/− [45], Il18 −/− [46,47] and Il18r1 −/− [47] mice, were shown to develop maturity-onset obesity, suggesting the involvement of age-related changes in the SNS in the phenotypes of these cytokine-deficient mice. It is possible that inflammatory cytokines are protective against age-related declines in β-adrenergic responsiveness.…”

“…While initial studies demonstrated that Il6 knockout (Il6 −/− ) mice display mature-onset obesity [16], subsequent studies did not verify this phenotype [17]. It is also unclear whether these mice display insulin resistance.…”
