Interleukin-1 Gene Cluster Polymorphisms in Sarcoidosis and Idiopathic Pulmonary Fibrosis

Abstract: Members of the interleukin-1 (IL-1) family are implicated in the pathogenesis of sarcoidosis and idiopathic pulmonary fibrosis (IPF). We have, therefore, performed a case-control study to investigate a plausible association between sarcoidosis and the polymorphisms in the IL-1alpha, IL-1beta, and IL-1 receptor antagonist (IL-1Ra) genes. Further, as a separate question, we explored whether the aforementioned genes of the IL-1 cluster are associated with IPF. Using PCR with sequence-specific primers, IL-1alpha -… Show more

“…In other studies the variable number of tandem repeats (VNTR) in intron 2 of IL1RN was investigated and found to be in linkage disequilibrium with the IL1RN + 2018 SNP. However, both a small Australian [7] and an independent Czech cohort [12] 1·3 (0·5-3·8)* 0·2 (0·1-0·3) Eosinophils (¥10 6 ) 0·8 (0·4-2·7)* 0·03 (0·01-0·08) *P < 0·05 compared to healthy controls. “…An IL1A single-nucleotide polymorphism (SNP), IL1A À889 C to T, has been associated with myasthenia gravis, 17 sarcoidosis, 18 age of onset of migraine 19 and juvenile rheumatoid arthritis. 20 Two SNPs in IL1B have reported genetic associations with disease.…”

“…IL1b and IL-1 receptor antagonists (IL-1RN or IL-1RA) both are very important members of the IL-1 family. 9 Both IL-1b and IL-1RN molecules bind to IL-1 receptors. IL-1b is a potent proinflammatory cytokine, while IL-1RN acts as an anti-inflammatory cytokine that competes with IL-1b for binding to IL-1 receptors without intrinsic effects.…”
