Abstract: Invasion of trophoblast cells is spatio-temporally regulated by various cytokines and growth factors. In pregnancy, complications like preeclampsia, shallow invasion of trophoblast cells and low amounts of epidermal growth factor (EGF) have been reported. In the present study, regulatory mechanisms associated with EGF-mediated invasion in HTR-8/SVneo trophoblastic cells have been delineated. Treatment of HTR-8/SVneo cells with EGF (10 ng/ml) led to eight fold increase (p < 0.05) in invasion. Increased invasion… Show more

“…Inhibition of ERK activation, such as by using the chemical inhibitor U0126, has been reported to downregulate the migration and invasion processes of multiple cell types. 22 , 23 Here, we also confirmed its similar anti-invasion role on MKN45 cells, which is consistent with the effect induced by PTPRF. Since MKN45 exhibits positive expression of EGFR and PDGFR proteins, 24 , 25 it is possible that PTPRF inhibits gastric adenocarcinoma progression by attenuating growth-factor effects.…”

“…Putative STAT1 and STAT3 binding sites in the promoter regions (5 kb upstream) of miR-92a-1-5p, using Genomatix MatInspector software were predicted (table S3 ) 17 . STAT1 and STAT3 are activated after EGF treatment of HTR-8/SVneo cells and play an important role in their increased invasion 18 . Additionally, STAT1 and STAT3 binding sites are present in the promoter region of miR-92a-1-5p.…”

“…TFs thus also control miRNA expression in the cell. EGF treatment of HTR-8/SVneo cells leads to STAT1 and STAT3 activation 18 ; and STAT1 and STAT3 in either homodimer form or heterodimer form act as transcription factors. As multiple binding sites for STAT1 and STAT3 were present in the 5 kb upstream promoter region of miR-92a-1-5p (table S3 ), effect of STAT1 & STAT3 silencing was determined on the expression of miR-92a-1-5p.…”

“…JAK/STAT3 signalling is indispensable for regulation of EVT proliferation and invasion capacity in response to cytokines and growth factors [145, 146]. Again, an interdependence between MAPK and JAK-STAT signalling pathways was found to be involved in EGF-mediated HTR-8/SVneo cell invasion [146].…”
