Abstract: Synergistic effects of BRAFV600E and TERT promoter mutations on the poor clinical outcomes in papillary thyroid cancer (PTC) have been demonstrated. The potential mechanism of this phenomenon has been proposed: MAPK pathway activation by the BRAFV600E mutation may upregulate E-twenty six (ETS) transcription factors, increasing TERT expression by binding to the ETS-binding site generated by the TERT promoter mutation; however, it has not yet been fully proven. This article provides transcriptomic insights into … Show more

“…In our recent study of PTC, we confirmed that TERT mRNA www.e-enm.org 521 expression was increased by the coexistence of BRAF V600E and TERT promoter mutations using RNA sequencing data from a relatively large number of PTC tumor samples (266 PTCs from TCGA and 65 PTCs from Seoul National University Hospital) [68]. The changes in the intracellular signaling pathways by BRAF V600E were further augmented by adding the TERT promoter mutation, and the pathways related to inflammation or adhesion molecules were upregulated by TERT expression, which is consistent with one of the non-canonical roles of TERT-namely, activating the NF-κB signaling pathway.…”

“…In a meta-analysis including 11 studies with 3,911 patients, PTC with concurrent BRAF and TERT promoter mutations were associated with increased tumor aggressiveness and had worst prognosis in comparison with PTCs harboring BRAF or TERT promoter mutation alone [81]. Recently, Song et al suggested the potential mechanism of synergistic effect of BRAF and TERT promoter mutations on cancer progression [82]. They explained that BRAF mutation activated MAPK pathway, and it upregulated E-twenty six (ETS) transcription factors.…”

“…Thus, our results provide preliminary evidence that GABPB2, which is typically expressed only at very low levels in GBM, could be targeted in conjunction with GABPB1L for increased treatment robustness. Multiple other transcription factors have also been linked to TERT regulation, both in normal and oncogenic contexts [53][54][55][56] . Such factors could play a role in basal expression of TERT off the mutant promoter, compensation for loss of GABPB1L, or both, and merit future study.…”
