Interaction of Bovine Serum Albumin with Chrysotile: Spectroscopic and Morphological Studies

Abstract: The biodurability of chrysotile fibers, which is related to their cytotoxicity and mutagenic responses, is strongly affected by the surface chemical adsorption of biological molecules. Natural chrysotile is a heterogeneous material in both structure and composition. The availability of synthetic stoichiometric chrysotile of constant structure and uniform morphology has allowed us to investigate its interaction with bovine serum albumin (BSA). By using transmission electron microscopy (TEM) and atomic force mic… Show more

“…The peak at 1090 cm −1 is attributed to the vibration modes of Si-O network that consists of Si-O-Si and O-Si-O, and the Si-O stretching peak which is vertical to the prolongation of the fiber, was undetected at 1024 cm −1 due to natural chrysotile tetrahedral sheets attacked by HCl. The peak at 617 cm −1 was identified as the terminal Si-OH deformation band, the Si-OSi bending peak was detected at 465 cm −1 (Wypych et al 2005;Wang et al 2006;Halma et al 2006;Falini et al 2006).…”

“…The geoinspired synthetic chrysotile fibres with stoichiometric composition Mg 3 Si 2 O 5 (OH) 4 , in contrast to the natural ones, do not exert any significant cytotoxic effect [15]. Moreover, the interaction of this synthetic chrysotile with bovine serum albumin (BSA) and human serum albumin (HSA) has been investigated [16][17][18].…”

“…From the second derivative of the bands, the actual half-width can be estimated, based on which a curve-fitting process was iterated to achieve the best fitted curves. The component bands were assigned on the basis of the literature data [16,[28][29][30][31][32][33].…”

“…For a series of studies concerning the interaction between drugs and protein, it is necessary to determinate the respective binding constant (K). So far, many studies have been carried out to investigate the interaction of proteins with drugs by fluorescence spectroscopy [5][6][7], FTIR [8,9] and CD spectroscopy [10,11]. Fluorescence techniques are one of the powerful tools in the study of interactions between drugs and plasma proteins in general and serum albumin in particular because of their high sensitivity, rapidity and ease of implementation.…”
