Integrating Molecular Networking and Biological Assays To Target the Isolation of a Cytotoxic Cyclic Octapeptide, Samoamide A, from an American Samoan Marine Cyanobacterium

Abstract: Integrating LC-MS/MS molecular networking and bioassay-guided fractionation enabled the targeted isolation of a new and bioactive cyclic octapeptide, samoamide A (1), from a sample of cf. Symploca sp. collected in American Samoa. The structure of 1 was established by detailed 1D and 2D NMR experiments, HRESIMS data, and chemical degradation/chromatographic (e.g., Marfey’s analysis) studies. Pure compound 1 was shown to have in vitro cytotoxic activity against several human cancer cell lines in both traditional… Show more

“…An excellent tool for known and unknown compounds is the GNPS (Global Natural Products Social molecular networking), where MS/MS spectra are uploaded and, through the MS/MS are grouped forming chemically similar clusters. Thereafter, the known compounds are recognized by GNPS and nodes of unknown compounds can be selected as the target of possible new metabolites [80,81,[84][85][86].…”

“…In a natural product molecular network, these clusters represent molecular families putatively synthesized by gene cluster families. The visualization of molecular networks in GNPS represents each spectrum as a node, and spectrum-to-spectrum alignments as edges (connections) between nodes (Naman et al, 2017).…”

“…3 The key is based on structurally related natural products that share similar tandem mass fragmentation patterns, and molecular families that tend to cluster together and can be visualized as a network. 2,4 This dereplication strategy has been widely used for the identication of marine and microorganism-derived natural products. 4,5 The marine environment represents approximately 70% of the earth's surface, and has a huge biodiversity, characterized by a wide chemical diversity of natural products.…”
