Abstract: The molecular features of necroptosis in cardiac ischemia-reperfusion (IR) injury have been extensively explored. However, there have been no studies investigating the physiological regulatory mechanisms of melatonin acting on necroptosis in cardiac IR injury. This study was designed to determine the role of necroptosis in microvascular IR injury, and investigate the contribution of melatonin in repressing necroptosis and preventing IR-mediated endothelial system collapse. Our results demonstrated that Ripk3 w… Show more

“…In the current study, we demonstrated that hyperglycemia‐mediated mitochondrial damage can be attributed to mitochondrial fission activation. Excessive mitochondrial fission produced numerous mitochondrial fragmentations, evoking ROS overproduction, mPTP opening, cyt‐c leakage, and caspase‐9 activation (Liu, Zeng, Li, Mehta, & Wang, ; Zhou et al, ; Zhou, Wang, Zhu, Hu, & Ren, ; Zhou et al, ). We also identified DUSP1 as the regulator of hyperglycemia‐mediated fission augmentation, because overexpression of DUSP1 repressed the formation of mitochondrial fragmentation.…”

“…After occlusion of the left anterior descending coronary artery using 7 to 0 silk suture (US Surgical Corp, Norwalk, Conn) for 45 minutes, the snare was released, and reperfusion was achieved for 4 hours (n = 6/group). Melatonin was initially dissolved in absolute ethanol and was subsequently diluted in sterile water to a final concentration of 0.5% ethanol according to our previous study . Melatonin (20 mg/kg) was intraperitoneally administered 24 hours before cardiac I/R injury.…”

“…Melatonin participates in many systems of the body, such as immune regulation, prevention of cancer metastasis, sleep regulation, and circadian rhythms, in addition to its hormone functions . Given its antioxidative, anti‐inflammatory, and antiapoptotic properties, melatonin is suggested to play a key role in attenuating reperfusion‐mediated myocardial damage . Moreover, the inhibitory effects of melatonin on mitochondrial fission have been extensively explored by our previous studies .…”

“…To explain the action of mitochondrial fragmentation, its inhibitor and activator were used. Mdivi-1, the inhibitor of mitochondrial fragmentation, was applied to hyperglycaemia-treated cells, which was used as the negative control group [41]. In MKP1-overexpressing cells, FCCP, an activator of mitochondrial fragmentation, was administered, which was used as the positive control group.…”
