Inhibition of dextran sulphate sodium (DSS)-induced colitis in mice by intracolonically administered antibodies against adhesion molecules (endothelial leucocyte adhesion molecule-1 (ELAM-1) or intercellular adhesion molecule-1 (ICAM-1))

Abstract: We examined the effect of intracolonic administration of anti-adhesion molecule antibodies on DSS-induced colitis in mice. Immunohistochemical staining in mice with colitis showed increased expression of ELAM-1 and ICAM-1 on endothelial cells of vessels in the lamina propria and submucosa at sites of inflamed lesions. Intracolonic administration of anti-ELAM-1 or anti-ICAM-1 antibody decreased bloody stools, anaemia, and histologically evident damage, as well as myeloperoxidase activity and IL-1beta content. “…Colitis severity was assessed by histopathological evaluation of the H and E-stained tissue sections as described above. Disease activity index (DAI) was calculated by determining body weight, stool consistency, and the presence of overt blood in stools or around the anus, substantially similar to that reported earlier [30] .…”

“…Body weight and disease activity index (DAI) were assessed on a daily basis. DAI was calculated as described earlier (19), combining weight loss, stool consistency, and stool blood content/ rectal bleeding. Mice were sacrificed at day 7 and colons were removed for further analysis.…”

“…Our group and others have found a difference in the expression of adhesion molecules in Crohn's disease compared with ulcerative colitis (Hemler, 1988;Malizia et al, 1991;Yacyshyn et al, 1994). Specifically, others have studied the role of ICAMs in gut inflammation (Wong et al, 1995;Hamamoto et al, 1999;Sans et al, 1999;Bendjelloul et al, 2000).…”

“…Several animal models have been developed to study IBD. In the past, many of these models have involved the mechanical induction of colitis with di-or trinitrobenzene sulfonic acid, dextran sulfate sodium (DSS), or acetic acid (Yamada et al, 1992;Wong et al, 1995;Hamamoto et al, 1999;Yoshida et al, 2001). More recently, gene knock-out, transgenics, leaky gut, and adoptive transfer rodent models have been created that develop a spontaneous and chronic form of inflammation involving the colon and other parts of the intestinal tract (MacDonald et al, 2000;Neurath, 2000).…”
