Infrared spectroscopy as a tool for discrimination between sensitive and multiresistant K562 cells

Abstract: Fourier transform infrared spectroscopy was performed on human leukemic daunorubicin-sensitive K562 cells and their multiresistant counterpart derived by selection. Statistical analysis, including variable reduction and linear discriminant analysis was performed on sensitive and multiresistant cells spectra in order to establish a diagnostic tool for multiresistant pattern. For each of the two methods of data reduction tested [genetic algorithm or principal component analysis (PCA)] discrimination between the … Show more

“…The solvent was slowly evaporated under a gentle stream of N 2 flow for 30 min as described previously [22,23,27]. Control cells (not treated with CLX) for both HT29 and SW620 were also grown and collected from five separate flasks.…”

“…As shown in Figure 5(a), the FT-IR spectra of the amide I band of the protein, assigned to the C=O of the peptide bond, are located in the 1700-1600 cm −1 region and the frequency of this peak is sensitive to protein secondary structure, while the amide II band, 1500-1570 cm −1 , is due to a combination of C-N stretching and N-H bending vibrations of the protein backbone [27,28]. The bands at 2955, 2872 and 2926 cm 1 are ascribed to CH 3 asymmetric, symmetric vibrations and CH 2 asymmetric vibrations, respectively.…”

“…26,29,30 In this study, the predictive model was applied to infrared images of 24 new test tumor sections (Table 1) from the 20 remaining tumors. Unidentified test spectra were analyzed by the PCA-LDA model that identified their tissue classes and colored them in accordance with their class color (dark blue color to represent tumor necrosis, yellow for viable tumor, blue for the fibrosis, green for liver parenchyma, and orange for liver parenchyma necrosis).…”
