Abstract: Remote ischemic conditioning (RIC) protects against sustained myocardial ischemia. Because of overlapping mechanisms, this protection may be altered by glyceryl trinitrate (GTN), which is commonly used in the treatment of patients with chronic ischemic heart disease. We investigated whether long-term GTN treatment modifies the protection by RIC in the rat myocardium and human endothelium. We studied infarct size (IS) in rat hearts subjected to global ischemia-reperfusion (I/R) in vitro and endothelial function… Show more

“…An unbalance between ROS production and defensive capacity of the cells leads to oxidative stress (Birben et al 2012;Tarantino et al 2011). Adhering to these hypotheses and that remote ischemic conditioning protects against endothelial I/R injury through ROS and NO-dependent mechanisms in the acute myocardial IRI model (Hauerslev et al 2018), our results showed that RIPerC provided protection against oxidative stress. Elevated MDA level, which has been frequently observed in animal models of renal I/R, is definitive feature of I/R-induced tissue damage (Sehirli et al 2003).…”

“…In addition, a series of experiments in which plasma from healthy volunteers subjected to RIPre was perfused through isolated mouse hearts, identified plasma nitrite as the cardioprotective agent [ 110 ]. Similarly, in a recent study by Hauerslev et al, scavengers of NO attenuated RIPre-induced protection in rat hearts [ 47 ]. Previously, it had been shown that either stimulation of cervical vagal nerves or perfusion with acetylcholine is associated with NO release in an isolated rabbit heart via the neuronal isoform of NO synthase [ 16 ].…”

“…Furthermore, prolonged exposure to GTN can induce tolerance. For example, 7 days of transdermal GTN exposure abolished pacing-induced preconditioning in conscious rabbits [ 35 ] and abrogated the effectiveness of remote ischaemic conditioning in rat myocardium and human volunteers [ 10 ]. Interestingly, however, direct myocardial protection with glyceryl trinitrate (GTN) may be preserved in the state of vascular nitrate tolerance [ 5 ].…”
