Infiltrating natural killer cells bind, lyse and increase chemotherapy efficacy in glioblastoma stem-like tumorospheres

Abstract: Glioblastomas remain the most lethal primary brain tumors. Natural killer (NK) cell-based therapy is a promising immunotherapeutic strategy in the treatment of glioblastomas, since these cells can select and lyse therapy-resistant glioblastoma stem-like cells (GSLCs). Immunotherapy with super-charged NK cells has a potential as antitumor approach since we found their efficiency to kill patient-derived GSLCs in 2D and 3D models, potentially reversing the immunosuppression also seen in the patients. In addition … Show more

“…NK cells can be genetically engineered to express CAR to increase their cytotoxicity function and to guide them toward tumors [ 184 ]. In glioblastoma, engineered NK-CAR cells targeting HER2, EGFR, and EGFRvIII were investigated in 3D tumors [ 152 ]. These 3D structures presented a dispersed morphology with an increase in the number of dead cells and in INF-γ, IL-6, and IL-8 secretion when co-cultured with the super-charged NK cells.…”

“…Advances in chemotherapy, radiation, and neurosurgical procedures have increased survival of patients with brain tumors, but curative treatments are not yet available to malignancies like glioblastoma due to their recurrence and/or progression. NK cell-based therapies have shown efficacy in pre-clinical studies making them a promising treatment against glioblastoma ( 31 , 32 ). NK cell-based immunotherapies provide an allogeneic “off-the-shelf” alternative to T cell therapy since they do not cause graft-versus-host disease.…”

“…In GBM, NK cells, identified by the CD56 marker, are present in perivascular tumor regions but are often functionally impaired in the peripheral blood of GBM patients [ 150 ]. This impairment influences their capability for tumor cell lysis, which is also modulated by surface molecules on glioblastoma stem-like cells (GSLCs), including CD44, CD54, MHC class I, and PD-L1.…”
