Infectious Bursal Disease Virus: Ribonucleoprotein Complexes of a Double-Stranded RNA Virus

Abstract: Genome-binding proteins with scaffolding and/or regulatory functions are common in living organisms and include histones in eukaryotic cells, histone-like proteins in some double-stranded DNA (dsDNA) viruses, and the nucleocapsid proteins of single-stranded RNA viruses. dsRNA viruses nevertheless lack these ribonucleoprotein (RNP) complexes and are characterized by sharing an icosahedral T=2 core involved in the metabolism and insulation of the dsRNA genome. The birnaviruses, with a bipartite dsRNA genome, con… Show more

“…6B), allowing translation of the GLV proteins and packaging into nascent, progeny GLV virions. This strategy for transcription inside the icosahedral capsid is thought to be common to all dsRNA viruses except possibly birnaviruses (67), which lack the "Tϭ2" capsid organization (68). The only solvent-accessible channels that span the GLV capsid appear to occur at each of the 12 5-fold axes, and though somewhat narrow (ca.…”

“…Segment A (3.3 kb) has two partially overlapping open reading frames. Open reading frame A-1 encodes a polyprotein that is autocatalytically cleaved to yield the immature outer capsid protein pVP2, the viral protease VP4 and the ribonucleoprotein VP3 (Sánchez & Rodriguez, 1999;Lejal et al, 2000;Da Costa et al, 2002;Luque et al, 2009). The pVP2 protein is further processed to yield the mature VP2, the immunodominant antigen of IBDV (Fahey et al, 1991).…”

“…Members of the families Birnaviridae are exceptions to the rule that all dsRNA viruses have a Tϭ1 core made of 60 dimers of the CP; birnaviruses have a single Tϭ13 shell (16,59) that surrounds a polyploid dsRNA genome (41) organized as a ribonucleoprotein complex (42).…”
