Abstract: The IL-23/T helper 17 (Th17) axis plays an important role in joint inflammation in ankylosing spondylitis (AS). Conventional CD4+ Th17 cells are a major source of IL-17A. IL-22 is another cytokine implicated in AS pathophysiology and is produced by Th17 and Th22 cells. In this study, we aimed to analyze conventional and non-conventional T cell subsets producing IL-17A and IL-22 in patients with AS. We thus evaluated the intracellular staining for IL-17A, IL-22, and IFN-γ in peripheral blood mononuclear cells o… Show more

“…In addition, IL-17 and granulocyte monocyte colony stimulating factor (GM-CSF) double producing γδ T cells were found to be increased in PB of AS patients when compared to RA patients and normal PB, similar to what was found with respect to CD4, CD8 and natural-killer (NK) cells [82]. Another study confirmed that double expressing IL-17 and GMCSF γδ as well as CD4, CD8, and ILC cells are found in synovium and increased in PB of AS patients, whereas there was a decreased percent of γδ T cells in their PB relative to normal [83]. Interestingly, in contrast to IL23R + IL-17 + Vδ1 + T cells that are expanded in AS, Vδ2 + γδ T cells were found to be increased in patients with AS who were receiving infliximab (anti TNFα) therapy, relative to those not receiving this drug and HC.…”

“…In Mr1 −/− NOD mice, exacerbation of diabetes development is associated with a loss of gut barrier integrity, allowing the translocation of bacterial compounds and subsequent dendritic cell activation in pancreatic lymph nodes 70 . Of note, a protective role of IL-17 has been established in several intestinal inflammation models 116 , and IL-22 + MAIT cells have been observed in the blood of patients with UC 18 and AS 84 .…”

“…Accordingly, total splenocytes from Mr1 −/− mice produce lower levels of IL-17 after anti-CD3 and anti-CD28 stimulation or IL-23 treatment. Human studies highlight a switch towards a T H 17-type profile of blood MAIT cells, with an increase in secretion of IL-17 in patients with CD 18 , UC 18,79 , ankylosing spondylitis (AS) 77 , obesity 16,73 , type 2 diabetes (T2D) 16 and cirrhosis 69 , and of IL-22 in patients with UC 18 and AS 84 . Analysis of MAIT cells in patients with MS has further shown increased expression of RORγt, CCR6 and IL-7R compared with healthy controls 118 .…”

“…In addition, MAIT cells were found to be involved in AS. Although the frequency of circulating MAIT cells was lower than in healthy controls, the proportion of IL-17+ MAIT cells in the blood of patients with AS was elevated [ 87 , 88 , 89 ]. Higher proportions of MAIT cells were found in the synovial fluid than in the circulation.…”
