For a better understanding of virus×host interactions, transmission electron microscopy was used to characterize the intrahaemocoelic infection of Anticarsia gemmatalis larval haemocytes by A. gemmatalis M nucleopolyhedrovirus (AgMNPV). At 12 h post-infection (h p.i.), we observed nuclear hypertrophy, budded virus assembling, and protrusion towards the cytoplasm, virion envelopment, and accumulation of fibrillar aggregates in the cytoplasm. Around 24 h p.i., fibrillar aggregates also appeared inside nuclei of infected cells. By 48 h p.i., virogenic stroma and polyhedra were visualised in nuclei and at 72 h p.i., widespread infection in haemocytes was observed. Cell remnants and free polyhedra were phagocytosed by granular haemocyte 1 and plasmatocytes. Entire cells were phagocytosed only by plasmatocytes. Necrosis of infected cells was quite common, suggesting a putative cytotoxic response. Granular haemocyte 1 presented a more exuberant protrusion of budded viruses in comparison to other haemocytes. All types of haemocytes were shown to be infected, and the intense virus replication in some of these cells reveals the importance of haemolymph for AgMNPV spread in its natural host, a critical factor for permissiveness.

SUMMARYThe uptake of two forms of Trichoplusia ni multiple nucleocapsid nuclear polyhedrosis virus by SpodopleraJrugiperda cells in culture has been evaluated. Subtle differences in the Mg 2+ and Ca 2+ requirements for the uptake of cell-released virus (CRV) and polyhedron-derived virus (PDV) exist; otherwise, both forms of the virus appeared similar in their mode of uptake. Entry occurs by fusion of the plasma membrane of the cell with the envelope surrounding the virus nucleocapsid at neutral pH. The receptor turnover for the virus probably occurs rapidly since saturation of receptor sites for the virus was not achieved. The binding and penetration of the virus, and the uncoating and transport of the viral genome to the nucleus is rapid. Uncoating occurs predominantly in the cytoplasm. The lower infectivity of PDV, compared with CRV, for cells in culture may happen because this form of the virus accumulates in cell vacuoles and is unable to proceed with infection.

Lepidopteran nucleopolyhedroviruses are members of the Baculoviridae and have been categorized as having two morphotypes of occluded virions: multiple nucleocapsids or single nucleocapsids within the virion envelope. Although it is a definitive characteristic of specific viruses, it appears to lack a defined genetic basis and is independent of virus phylogeny. This review summarizes the factors that appear to influence this trait and the role that it may play in virus biology.
