In vitro grown human ovarian follicles from cancer patients support oocyte growth

Abstract: Our data support the notion that human follicle development can be achieved in vitro in a bio-engineered culture system. More studies are required to investigate whether the fully sized oocytes obtained from in vitro grown follicle are competent to resume meiosis and be fertilized.

“…Until now, however, the production of live offspring from primordial and pre-antral follicles from cryopreserved/thawed ovarian tissue and cultured to mature in vitro has only been reported in immature mice (Liu et al 2001, O'Brien et al 2003, Hasegawa et al 2006. No live births have previously been reported using adult mouse ovaries, and although development of follicles to the antral stage has been observed in human ovarian tissue cultured in vitro (Isachenko et al 2003, Xu et al 2009, there have been no reports assessing the competency of the oocytes obtained from these in vitro grown follicles.…”

“…Isolated primordial follicles were encapsulated in either 0.5 % or 2 % alginate (Sigma-Aldrich) in phosphatebuffered saline (PBS) as previously described [36]. Briefly, groups of follicles were transferred to alginate, and beads were formed by pipetting drops of alginate into the cross-linking solution (50 mM CaCl 2 and 140 mM NaCl).…”

“…To overcome the limitations of ovarian tissue transplantation, an alternative fertility preservation approach, in vitro follicle growth (IVFG), is actively being developed [35,36]. In this method, ovarian tissue or follicles isolated from ovarian tissue can be grown in vitro to produce mature gametes that could ultimately be used in standard ART procedures.…”

“…Our basal 3D culture system employs the biomaterial alginate, which has been used successfully in the culture of murine, monkey, and human immature follicles [7][8][9][10][11]. Compared with 2D culture systems, follicles cultured in the alginate hydrogel have similar growth trends in follicle diameter and steroid production, yet produce a significantly greater percentage of mature oocytes.…”
