Micropropagation protocol of Oriental Hybrid Lilium cv. Ravenna was developed using bulb scale segments (Basal and Tip) as explants. Surface sterilization of healthy bulb scales with carbendazim 200 ppm for 30 min, then 0.1 percent mercuric chloride for 10 min, then 70% ethyl alcohol for 30 s was superior to all other treatments in recording highest culture asepsis (77.08%) and higher explant survival (86.12%). Explant survival was higher in basal segments (88.54%) compared to tip segments (85.52%). Highest culture establishment was recorded in basal scale segments (68.26%) followed by tip scale segments (55.21%). MS medium augmented with 0.50 mgl −1 Naphthalene acetic acid and 2.0 mgl −1 . 6-Benzylamino Purine recorded maximum culture establishment (76.17%), highest bulblet number/explant (5.52) with maximum length of shoots (2.20 cm) and number of leaves (3.39). This treatment combination of growth regulators resulted in highest shoot proliferation (83.33%) along with maximum shoot number (2.41explant −1 ), shoot length (2.35 cm) and leaf number (5.44) of micro shoots during proliferation stage. Rooting of explants was superior with Indole-3-butyric acid compared to Naphthalene acetic acid. Highest rooting of 92.71% along with maximum number of primary roots shoot −1 (12.06), maximum primary root length (3.17 cm) was documented in Murashige and Skoog medium added with Indole-3-butyric acid 1.50 mgl −1 with best ex vitro survival rate (98.96%) of rooted plantlets during primary hardening in perlite + vermiculite (1:1) mixture.

Aim: Developing a protocol for mass multiplication of Lilium (Lilium longiflorum) bulbs via indirect organogenesis and somatic embryogenesis for in-vitro plant regeneration and ISSR marker-based mapping to assess the fitness of true to type. Methodology: To induce organogenic callus, six combinations of 2,4-D and BAP were tested; while nine combinations of picloram and NAA were used for somatic embryogenesis. NAA and IBA were tested for root induction. The clonal fidelity of the in-vitro regenerated plantlets from both calluses were tested using polymerase chain reaction (PCR)-based ISSR markers analysis. All experiments were arranged in a complete randomized block design and replicated three times. Results: Two methods (direct and indirect organogenesis) were investigated to assess the best callus mediated plant regeneration for Lilium bulb multiplication. The maximum organogenic callus induction and the highest regeneration percent was recorded with BAP (0.5 mg l-1) + 2,4-D (3.0 mg l-1). However, MS medium containing 0.50 mg l-1 picloram and 0.20 mg l-1 NAA was found best for initiation of embryogenic callus and proliferation and was found superior over direct organogenesis. Clonal fidelity was assessed through ISSR markers comparing the mother plant and regenerated plantlets. Interpretation: Both organogenic callus andembryogenic callus are capable of developing true to type in-vitro plants and can be explored for mass multiplication of Lilium bulbs. Embryogenic callus can further be utilized in liquid suspension based bioreactor system. The present protocol has potential applications in micropropagation and genetic transformation studies in other Lilium spp.

Background: Lily is one of the most important commercial cut flower species and one of the three major bulb crops in the commercial market. The aim was to assess and establish a suitable protocol for increasing the number of Lilium bulblets and enlarge their size. Methodology: Different plant growth regulators (TDZ, 2,4-D, 2ip, and picloram) at different concentrations for direct and indirect micropropagation were used. Also, the effect of various concentrations of sucrose alone or with growth inhibitor on bulblet formation during three repeated subcultures was examined with anatomical study. Results: For direct shoot organogenesis, culturing leaf explant on MS culture medium containing TDZ (0.5 mg/l) + 2,4-D (10 mg/l) led to the highest number of shootlets after 1 month. For indirect micropropagation, MS was supplemented with 2ip (0.5 mg/l) + picloram (5 mg/l)-induced callus tissues which were differentiated to embryos then weak shoots after 3 months. For more proliferation, the highest numbers of shootlets, leaves, and bulb scales were observed on MS culture medium supplemented with 1.0 mg/l of BA plus 0.2 mg/l NAA. Using 60 g/l of sucrose plus 3 or 6 mg/l of paclobutrazol led to the highest number of bulblets/explant for the third subculture. The highest bulblet diameters were recorded with 120 g/l sucrose plus 6 or 3 mg/l paclobutrazol. Conclusion: This study had reached to optimize a suitable protocol for direct and indirect micropropagation of Lilium orientalis cv. "Starfighter" bulbs and increase the number and size of bulbs that can be beneficial for increasing the production and reduce their price. This study will help the producers for commercial purposes.
