Improved method for chick whole‐embryo culture using a filter paper carrier

Abstract: We describe a simple method of chick whole-embryo culture, which uses a filter paper carrier to hold the early blastoderm and vitelline membranes under tension while the embryo grows on a substratum of agar-albumen. This is a quick and efficient means of setting up cultures of chick embryos beginning at pre-primitive streak stages to stage 10 (stages X--XIV, Eyal-Giladi and Kochav [1976] Dev Biol 49:321-337; stages 1--10, Hamburger and Hamilton [1951] J Morphol 88:49--92). This is an improvement on the origina… Show more

“…The embryo culture was prepared as described by (Chapman et al, 2001). The embryo was first turned upside down (ventrally) and the endoderm above the area of operation was removed then a drop of 0.05% pancreatin (Gibco-BRL) in calcium free PBS was applied for 1 min.…”

“…Chick embryos were isolated and placed in a modified New culture for in vitro manipulation (Chapman et al, 2001). Briefly, eggs were placed into a 10 cm plate, and the albumen was scraped off of the embryonic region with a razor blade.…”

“…We utilised a chick embryo culture method that allows embryos to be maintained upon a semi-solid base for up to 24 h and facilitates manipulation and treatment with exogenous reagents (Chapman et al 2001). This technique was used in conjunction with the plant steroidal alkaloid, cyclopamine (obtained from Toronto Research Chemical, Canada), a potent inhibitor of Shh signalling (Taipale et al 2000).…”

“…Chicken IGF-I and -II were also included to establish where there was overlap of expression with receptor and ligand. Finally, we employed cyclopamine treatment of embryos in EASY cultures (Chapman et al 2001) to establish whether the action of IGFBP-5 lies downstream in a signalling pathway from the morphogen Sonic Hedgehog (Shh) during early chick embryogenesis.…”
