Objective: The study’s goal was to ascertain the contraceptive effects of Aqueous extract of Carica papaya on female rats by assessing changes in the body weight, estrous cycle, serum progesterone level and the cyto-architecture of the Utero-ovarian tissue. Methods: We used twenty (20) healthy young Adult Female Albino rats. The study ran for 7 and 21 days, respectively. Each study group has their Experimental (treated 200mg/kg aqueous extract of Carica papaya seed extract) and Control group (n=5). We determined daily the phases and frequencies of the estrous cycles of the rats during the administration of the extract. We processed the utero-ovarian tissue for histological analysis, and we assessed serum progesterone level and the oestrus cycle pattern. Results: There was a significant increase in body and Ovarian weights after 21 days of treatment when compared to controls and those treated for 7 days. However, uterine weight reduced significantly ( p <0.05), serum progesterone level decreased ( p <0.05) in the treated rats, mostly in those submitted to 21 day-treatments; the ovary showed marked degeneration of the theca cells, granulosa and corpus luteum, and loss of mucin granules in the uterine tissues. Carica papaya administered for 7 and 21 days caused the animals to have more proestrus and diestrus phases as compared to the control animals. The estrous cycle became irregular, with prolonged diestrous and proestrus phase. Conclusion: The aqueous extract of Carica papaya seeds caused antifertility, anti-implantation, by a reduction in progesterone level, disruption of oestrus pattern and histological alteration of utero-ovarian tissue.

Penelitian ini bertujuan untuk menganalisis respon glomerulus ren mencit (M. musculus) terhadap pemberian senyawa antifertilitas dari esktrak air biji pepaya (Carica papaya L.). Penelitian ini merupakan penelitian eksperimental yang dibagi dalam 5 kelompok perlakuan dengan 3 ulangan. K(-) bahan uji berupa akuades, K(+) bahan uji berupa pil kontrasepsi dosis 0.2 mg/ml, P1 sampai P3 berupa bahan uji ekstrak air biji pepaya dosis masing-masing 1.4; 3.5; dan 7 mg/ml. Perlakuan diberikan secara oral selama 21 hari. Pengambilan sampel dilakukan dengan cara dislokasi leher setelah paparan bahan uji berakhir. Ren diisolasi dengan cara membedah dari abdomen bagian bawah. Ren selanjutnya, ditimbang dan dibuat sediaan histologis dengan pewarnaan HE dan tebal sayatan 8 mikron. Pengukuran diameter dilakukan pada setiap sayatan, masing-masing dengan 5 bidang pandang. Hasil penelitian dianalisa dengan menggunakan Analysis of Variance (ANOVA) pada taraf kepercayaan 95%. Hasil analisis berat ren kelompok perlakuan menunjukkan tidak berbeda bermakna (p>0.05) dengan kelompok kontrol. Diameter glomerulus kelompok perlakuan menunjukkan perbedaan bermakna (p<0.05) dari kelompok kontrol. Kesimpulan penelitian ini adalah paparan ekstrak air biji pepaya selama 21 hari berpotensi menyebabkan kerusakan ren, ditunjukkan dengan peningkatan ukuran diameter glomerulus.

Protection of plant extracts to prevent their degradation before uptake in the gastrointestinal tract could enhance short-term efficacy but would not enhance the longevity of their effects. Identification and further testing of the specific chemicals responsible for reproductive effects would be beneficial. The adoption of a standard protocol for treatment and assessment of the inhibitory effects of potential control agents on reproductive function in rodents is essential. Treatment with higher concentrations of extracts in conjunction with other extracts or with other chemosterilants could have potential complementary effects and lead to more rapid and permanent changes in ovarian function. An orally delivered agent(s) that causes major depletion of all follicle types, and particularly of non-regenerating primordial follicles, could be an ideal fertility control product and serve as an additional tool for population control of pest rodents.
