Immunoprotection in goats against Haemonchus contortus after immunization with cysteine protease enriched protein fractions

Abstract: -Haemonchus cysteine proteases, because of their apparent critical function in worm physiology, are considered important candidates in the immunological control of haemonchosis in sheep. Only limited information is, however, available on the immunoprotective properties of these molecules in goats. In the present study cysteine proteases of Haemonchus contortus adult worms isolated from a goat strain (Gran Canaria, Spain) were enriched by affinity chromatography and evaluated as immunoprotective antigens agains… Show more

“…79 Cysteine protease-enriched protein fractions of H. contortus adult worms were found to confer protection in goats against haemonchosis based on significant reductions in FEC output and worm burdens in immunized compared to nonimmunized animals. 81 Moreover, the cysteine protease-enriched protein fractions were found to have a cross-protective effect against H. contortus in both sheep and goats. 82 Fractions purified from sheep-adapted strains had a similar cross-protective effect with the fractions purified from goat-adapted H. contortus strains.…”

“…This time, the proteins were eluted from a thiol sepharose column using 25 mM cysteine followed by 25 mM Dl‐dithiothreitol and this preparation used to vaccinate sheep. Cysteine protease‐enriched protein fractions of H. contortus adult worms were found to confer protection in goats against haemonchosis based on significant reductions in FEC output and worm burdens in immunized compared to nonimmunized animals . Moreover, the cysteine protease‐enriched protein fractions were found to have a cross‐protective effect against H. contortus in both sheep and goats .…”

“…In a further experiment, this immunogen was subfractionated through Q-Sepharose anion exchange chromatography and a group injected with a resultant cysteine protease enriched fraction had a reduction of 80% in cumulative faecal egg output compared to controls [16]. A similar thiol-enriched fraction was tested in goats, with 89% and 68% in egg and worm reduction, respectively [25]. These experiments above used Freunds or QuilA as adjuvants, in contrast to the aluminium hydroxide employed in ours.…”
