Immunomodulatory effects of licochalcone A on experimental autoimmune encephalomyelitis

Abstract: LicoA possesses immunomodulatory effects on H2 O2 , NO, IFN-γ, TNF-α and IL-17 production in cells from EAE mice. It is suggested that LicoA acts on the mechanism of development of EAE by IFN-γ, IL-17 and TNF-α inhibition, modulating the immune response on both Th1 and Th17 cells.

“…This protocol results in a chronic demyelinating disease predominantly driven by CD4 + T cell-mediated immunity, especially Th1 and Th17 cells, pointed as the main immunopathogenic cells in this model [6,7]. Th1 cells, stimulated by IL-12 or IFN-γ, secrete IFN-γ and TNF-α that have been considered primarily responsible for the induction of the inflammatory process in CNS during the development of MS and EAE [8].…”

“…Briefly, mice received a subcutaneous injection at the tail base with 100 µg of MOG 35-55 (≥ 97 % purity; Sigma-Aldrich) per animal emulsified in CFA (Sigma-Aldrich) (v/v) containing 400 µg of attenuated Mycobacterium tuberculosis H37RA (Difco). Immediately thereafter, and again 48 h later, the mice received an intraperitoneal (i. p.) injection of 300 ng/animal of pertussis toxin (Sigma-Aldrich) [6]. Non-immunized mice were used as controls.…”

“…Chalcone, (E)-1,3-diphenyl-2-propene-1-one, has been reported as being a precursor in flavonoid synthesis [1]. Similar to flavonoids, chalcones have already been documented as exhibiting several biological activities, including antioxidant [2], anti-inflammatory [3], antibacterial and antifungal [4], antiprotozooal [5], immunomodulatory [6], and antiangiogenic [7,8] effects. Furthermore, antiproliferative/antitumor activities of chalcones have also been intensively studied [9][10][11][12][13][14].…”
