Immunological studies of Escherichia coli mutants lacking one or two ribosomal proteins

Abstract: A battery of immunological tests were used to investigate mutants which had been determined as lacking one or two ribosomal proteins on the basis of two-dimensional polyacrylamide gels. Proteins which were confirmed as missing from the ribosome in one or more mutants were large subunit proteins L1, L15, L19, L24, L27, L28, L30 and L33 and small subunit proteins S1, S9, S17 and S20. Cross-reacting material (CRM) was also absent from the post-ribosomal supernatant except in the case of protein S1. Since mutants … Show more

“…AM111, an erythromycin-independent revertant of AM, lacks ribosomal proteins S17 and L29 (21,22). AM111-5, selected as a faster-growing spontaneous derivative of AM111, lacks L29 alone (23). These observations, together with ours presented here, suggest that BSC and MC at least of etiolated maize leaves contain two different populations of functional chloroplast ribosomes with respect to L29: one class with and the other without L29.…”

“…The strong growth phenotype of ⌬S9 is consistent with a partial defect in ribosome assembly. Although Dabbs and coworkers (12)(13)(14) have previously reported S9 and S13 deletion strains, the method of Link et al (15) provided a means to create precise chromosomal deletions. We did not attempt to generate an S13 deletion strain.…”

“…By contrast, omission of S13 in reconstitutions carried out with purified components yielded particles whose sedimentation was indistinguishable from WT 30S subunits, but slightly defective in functional tests (8). In early genetic screens, Dabbs and coworkers reported deletions of both S9 (12,13) and S13 (14). The S9 deletion was obtained in a search for dependence on kasugamycin, an antibiotic that inhibits binding of fMet-tRNA fMet during initiation.…”

“…Since E. coli mutants lacking L27 are viable, albeit with severe impairment of growth, L27 is not absolutely essential for ribosome assembly and function (7). In contrast to this, we have classified MRP7 as an essential protein on the basis of our characterization of MRP7 null mutants, which are completely devoid of mitochondrial protein synthesis.…”
