Immunohistochemical location of the p75 neurotrophin receptor (p75NTR) in oral leukoplakia and oral squamous cell carcinoma

Abstract: These results suggest that p75NTR is expressed in undifferentiated cell populations in OL and OSCC. Furthermore, p75NTR is possibly involved in invasion and poor prognosis in OSCC.

“…71,72,73,74 One study showed that the labeling index of p75 was similar among normal and oral leukoplakia samples, whereas increased p75 expression correlated with a worse OSCC tumor grade. 75 Another group showed that K15 was down-regulated in oral lichen planus, while α6-integrin, β1-integrin and melanoma chondroitin sulphate proteoglycan were upregulated in both oral lichen planus and hyperkeratotic oral lesions. 76 Others also reported that oral dysplasias had decreased expression of p63 (ref.…”

“…Additionally, several studies have attempted to correlate putative CSC markers with patient prognosis and survival, showing that tumors expressing higher levels of certain CSC markers tend to have poorer outcomes. 72,75,78,79,80,81 One caveat is that unless serial transplantation assays have shown that the stem cell markers being studied actually label CSCs, it is difficult to draw firm conclusions about the identity and function of positively staining cells within OSCCs. This is true even for stem cell markers that have already been shown to label normal OESCs, since these cells may not perform similar functions within tumors.…”

“…According to the results of a scratch assay, p75 NTR+ cells possess migratory ability. Kiyosue et al (12) reported a significant association between the magnitude of p75 NTR positivity and the mode of tumor invasion. Furthermore, Soland et al (46) reported that the average risk of local recurrence was increased by ~17-fold in OSCC when p75 NTR expression was positive, and marked tumor cell dissociation was detected at the invasive front.…”

“…It has previously been reported that the p75 neurotrophin receptor (p75 NTR ) may be involved in the invasion and poor prognosis of OSCC (12). As a member of the tumor necrosis factor superfamily, p75 NTR is a 75-kDa cell-surface receptor glycoprotein (13,14), which is involved in diverse cellular responses, including cell proliferation and survival, and apoptosis in neural and non-neural tissues (15,16) via unique pathways (17,18) or activation of the intrinsic caspase pathway (19).…”

“…TrkA is expressed in basal and parabasal mucosal and gingival epithelial cells, with weaker expression in spinous and granular cell layers, while p75 neurotrophin receptor (p75 NTR ) only occurs in basal cell layers (Figure 5) [14]. p75 NTR is also expressed in oral squamous cell carcinomas (OSCCs) [16,17] and can be used as a prognostic marker together with the pattern of invasion. We found that high expression of p75 NTR in the central, superficial and invasive parts of the tumor was a marker for recurrence.…”
