Abstract: Gamma interferon, tumor necrosis factor alpha, interleukin-4 (IL-4), and IL-10 levels in supernatants and serum anti-PT IgG levels were determined using enzyme-linked immunosorbent assay (ELISA). The net percentage of CD3؉ blasts in cultures with B. pertussis in the group vaccinated with wP was higher than that in the group vaccinated with the wP low vaccine (medians of 6.2% for the wP vaccine and 3.9% for the wP low vaccine; P ‫؍‬ 0.029). The frequencies of proliferating CD4 ؉ , CD8 ؉ , and ␥␦ ؉ cells, cytoki… Show more

“…Our previous results showed that only 0.4% of healthy infants failed to mount seroprotection to hepatitis B 1 month after the completion of the three-dose vaccination schedule (6). In addition, the frequencies of subjects protected against tetanus and diphtheria vaccinated with three doses of DTP were 98.2 and 95.5%, respectively (38).…”

“…In particular, Toll-like receptor 4 (TLR4) in concert with CD4-positive type 1 T-helper cells (Th-1) and interleukin-17 (IL-17)-producing T-helper cells (Th-17) appear to play a central role in mediating protective immunity after whole-cell pertussis vaccination (408,411), whereas Th-1 cells alone mediate the protective immune response after recovery from B. pertussis infection (409,411). In infants, conventional whole-cell pertussis vaccines elicited a higher level of CD3 T-lymphocyte proliferation than did the low-lipopolysaccharide-content whole-cell vaccine (412). Other molecules playing a role in protection against pertussis include interferon gamma, which appears to protect against systemic dissemination (413), IL-12, which induces Th-1 cells (414), and tumor necrosis factor alpha (TNF-␣), which enhances macrophage phagocytosis of B. pertussis (415).…”

“…Still, new developments are being attempted in Brazil to further reduce reactogenicity problems, such as the development of a new vaccine that has reduced LPS content (wP low -for wP with low endotoxin level) after an organic solvent extraction, which was incorporated into the manufacturing process. This new vaccine was shown to be safe and to induce similar immune responses in a phase I clinical trial and is a good candidate for replacing the classical wP (58). Since wP will continue to be given at 2, 4, and 6 months of age as part of the triple bacterial vaccine -diphtheria, tetanus and pertussis (DTP) -, it could serve as an adjuvant for co-administered antigens.…”
