Immunochemical and biological characterization of outer membrane proteins of Porphyromonas endodontalis

Abstract: Outer membrane proteins (OMP) of Porphyromonas endodontalis HG 370 (ATCC 35406) were prepared from the cell envelope fraction of the organisms. The cell envelope that had been obtained by sonication of the whole cells was extracted in 2% lithium dodecyl sulfate and then successively chromatographed with Sephacryl S-200 HR and DEAE-Sepharose Fast Flow. Two OMP fractions, OMP-I and OMP-II, were obtained, and their immunochemical properties and induction of specific antibodies were examined. The OMP-I preparation… Show more

“…IgG is more stable than IgA, thus making it a more appealing antibody for clarifying a history of infection. In patients with ELs, the major immunoglobulin against P. endodontalis antigens produced at the site is IgG (Ogawa et al 1992). Therefore, ELs might lack the nature of repeated or recent antigen exposure, which is required for elevated P. endodontalis IgA levels.…”

“…A 41-kDa OmpA homolog has been identified in B. fragilis (30). P. endodontalis also has OmpA-like porins (31). These OmpA homologs were reported to have pore-forming activity.…”

“…Specific humoral immune response against several bacteria from the infected root canal has been investigated by the enzyme-linked immunosorbent assay (ELISA) method using crude bacterial components as antigens (Kettering et al 1991). Monoclonal and oligoclonal immunoglobulins were detected in periapical lesions (Matsumoto 1985) and periapical antibody-secreting cells against specific antigens of endodontopathic bacteria have been investigated by using enzyme-linked immunospot (ELISPOT) assay (Ogawa et al 1992). This evidence supports the possibility that`periapical lesion relevant' plasma cells may arise via selective homing and/or by the clonal proliferation and differentiation of specific B cells in this region.…”
