Immunoactivating Peptide P4 Augments Alveolar Macrophage Phagocytosis in Two Diverse Human Populations

Abstract: New treatment strategies are urgently needed to overcome early mortality in acute bacterial infections. Previous studies have shown that administration of a novel immunoactivating peptide (P4) alongside passive immunotherapy prevents the onset of septicemia and rescues mice from lethal invasive disease models of pneumococcal pneumonia and sepsis. In this study, using two diverse populations of adult volunteers, we determined whether P4 treatment of human alveolar macrophages would upregulate phagocytic killing… Show more

“…In addition to its role in metal acquisition, the P4 region (residues 251-278) of PsaA is immunogenic [13][14][15]. The P4 region has also been implicated in mediating PsaA adhesion to nasopharyngeal epithelial cells [11][12][13][14][15][16], although the in vivo relevance of this adhesin property remains contentious. The reduced efficacy of current vaccine regimes, together with the spread of multidrug resistant strains, highlights an urgent need to develop new therapeutic strategies for the management of pneumococcal infections [17].…”

“…bacterial killing using this control peptide so pragmatically used DEPC water as the vehicle control for the remainder of the study, as per previously published work (14,15). For the whole blood phagocytosis assay we noted convergence of results after 60 minutes incubation between P4 peptide and vehicle control stimulated samples.…”
