Immune Activation by Epidermal Growth Factor Receptor–Specific Monoclonal Antibody Therapy for Head and Neck Cancer

Abstract: To determine if the epidermal growth factor receptor (EGFR)-specific monoclonal antibodies (mAbs) cetuximab or panitumumab mediate in vitro immune activation against squamous cell carcinoma of the head and neck (SCCHN) cell lines.

“…The H1975 cells remained viable in the presence of all antibodies and antibody containing AuNPs (Supporting Information, Figure S1) 72 h post-treatment. The results are consistent with previous reports that show no in vitro cell death upon exposure of H1975 lung cancer cells to cetuximab or panitumumab and reveal that multiple antibodies on the surface of the AuNPs also do not produce any antiproliferative effects in vitro.…”

Exaggerated anticipatory anxiety is common in social anxiety disorder (SAD). Neuroimaging studies have revealed altered neural activity in response to social stimuli in SAD, but fewer studies have examined neural activity during anticipation of feared social stimuli in SAD. The current study examined the time course and magnitude of activity in threat processing brain regions during speech anticipation in socially anxious individuals and healthy controls (HC). Method Participants (SAD n = 58; HC n = 16) underwent functional magnetic resonance imaging (fMRI) during which they completed a 90s control anticipation task and 90s speech anticipation task.

“…In the present study, we extended our previous observations that cetuximab mediates cell-dependent cytotoxicity against SCCHN cells [11]. We found that the expression of an Fc γ R IIIa-158 V allele correlates strongly with ADCC activity, NK cell activation, degranulation and secretion of several cytokines with anti-tumor activity.…”

“…Indeed, panitumumab has been shown to bind the EGFR with an 8-fold greater affinity than cetuximab (23). Based on our prior studies (9) and this study using HNSCC cells, we observed that both panitumumab and cetuximab bind to EGFR and HNSCC cells with similar dilution curves and, importantly, both mAbs appeared to inhibit EGFR internalization and ligand-induced activation at similar doses. Thus, EGFR binding affinity is of uncertain clinical significance.…”
