Abstract: SYNOPSIS In recent years, genome-wide association studies (GWAS) all identified polymorphisms located near the gene encoding the interferon-lambda beta subunit (also known as IL28B) that turned out to be the best predictor of response to pegylated interferon plus ribavirin for chronic HCV genotype 1 infection. This association was extended to spontaneous clearance of HCV, suggesting shared mechanisms of both treatment and natural control of this virus. African-Americans were discovered to harbor the favorable … Show more

“…This is a systematic review and meta‐analysis of studies associating host genetic polymorphisms with spontaneous or treatment‐induced HCV clearance between 1989 and 2013. While previous reviews were not systematic, focused on a specific marker, gene or group of genes, and were limited to a viral genotype or ethnic group, this review includes all genetic determinants of HCV clearance without limitations. To our knowledge, this is the most comprehensive review on this topic so far.…”

“…In a prospective international cohort study of injectable drug users with acute HCV infection, HCV RNA levels 12 months following infection were independently associated with male gender, IL28B CC rs12979860 genotype and HCV genotype 1 [24]. The importance of these results remains to be elucidated, as some studies have refuted the effect of HCV RNA loads on disease progression, but it is probable that high levels of HCV RNA may have an impact on immunological response and hepatic inflammation especially in advanced liver injury [25,26].…”

“…Knowledge on the molecular pathways of nutrients, according to the genetic identity of the patients, will enable the application of the most optimal treatment for each patient with AMD. Genetic profiling would likely contribute to our understanding of AMD and potentially provide more effective management of the disease, such as the case in HIV [89,90] and HCV [91,92] infection.…”
