IgH Class Switch Recombination to IgG1 in DNA-PKcs-Deficient B Cells

Abstract: To assess the role of the DNA-PKcs nonhomologous DNA end-joining (NHEJ) protein in Ig heavy chain class switch recombination (CSR), we assayed CSR ability of DNA-PKcs-deficient (DP-T) B cells generated via complementation of DP-T mice with Ig heavy chain and light chain knock-in transgenes (DP-T/HC/LC mice). DP-T/HC/LC mice were severely deficient for all serum IgH isotypes except IgM and, unexpectedly, IgG1. Upon appropriate stimulation, DP-T/HC/LC B cells showed normal proliferation, germline C(H) gene trans… Show more

“…DSB intermediates in the switch reaction are repaired by a nonhomologous end-joining (NHEJ) pathway requiring Ku80/Ku70 (refs 9, 16) and partially dependent on DNAPKcs 17,18 . To determine whether Ku80 is also required for c-myc-Igh translocations we overexpressed AID by retroviral transduction in immunoglobulin transgenic Ku80 −/− B cells stimulated with LPS and IL-4 (Ku80 −/− HL) 9 .…”

“…DNA-PK is required for proper B-and T-cell antigen receptor rearrangement, and DNA-PK CS Ϫ/Ϫ mice develop severe combined immunodeficiency (30). Furthermore, Alt and colleagues have recently described a role for DNA-PK CS in immunoglobulin (Ig) class switching to the murine IgG2a isotype, an antibody subclass traditionally associated with cellmediated immune responses (39).…”

“…Ku70-and Ku80-deficient B cells have severely impaired CSR (Casellas et al, 1998;Manis et al, 1998;Reina-San-Martin et al, 2003), but this defect could result from a reduced cellular proliferative capacity or increased apoptosis of B cells rather than a CSR defect per se (Manis et al, 1998;Reina-SanMartin et al, 2003). Conflicting results were reported concerning the role of DNA-PKcs in CSR (Bosma et al, 2002;Manis et al, 2002a;Cook et al, 2003;Kiefer et al, 2007). Finally, Artemis deficiency does not seem to have any impact on CSR in such monoclonal B-cell mice (Rooney et al, 2005).…”
