Identification of Hippophae species (Shaji) through DNA barcodes

Abstract: BackgroundThe morphological identification of different Hippophae species (Shaji) was difficult. This study aims to discriminate between medicinal and non-medicinal Hippophae species by DNA barcodes, the ITS2, psbA-trnH, and a combination of ITS2 and psbA-trnH (ITS2 + psbA-trnH).MethodsDNA was extracted from the dried fruit samples. Primer pairs ITS2F/3R for ITS2 and psbAF/trnHR for psbA-trnH were used for PCR amplification. The purified PCR products were bidirectionally sequenced. Genetic distances were calcu… Show more

“…Currently, the Nrf2/ARE signaling pathway appears to be the most important endogenous antioxidant stress pathway [65,66]. Furthermore, Nrf2 is pivotal in the host defense mechanism, and numerous key phase II enzymes, such as NQO1 and HO-1 [67], are controlled by Nrf2 transcription factor under ROS conditions [4].…”

“…Plant metabolomics has recently attracted considerable attention because the method focuses on the holistic characteristics, and has been developed as an important approach for the modern research of (medicinal) plants. LC/GC-MS and 1 H NMR are the most commonly used analytical strategies in metabolomics 22 26 . Compared with the two chromatographic techniques, 1 H NMR based metabolomics is considered as the most promising analytical tool because the method allows simultaneously detection of primary and secondary metabolites in a single run, and produces a non-biased abundant metabolic profile.…”

“…The internal transcribed spacer 2 (ITS2) region has been proposed as a candidate DNA barcode for medicinal plants Han et al, 2016;Xiang et al, 2015;Yao et al, 2010). At present, the DNA barcoding technique has been successfully used to identify traditional Chinese medicines from adulterants and closely related species (Hu et al, 2015;Liu et al, 2015;Pang et al, 2013;Pang et al, 2011), such as Corni Fructus , Aristolochiaceous herbs (Wu et al, 2015), Ganoderma species (Liao et al, 2015), and Rhodiola species (Xin et al, 2015). Therefore, this study used the ITS2 region to distinguish Fritillariae bulbus from other species and adulterants.…”
