“…For the generation of bovine monocyte‐derived DC, PBMC were also derived as described above, and DC were generated from PBMC as described elsewhere 27 but with minor modifications. PBMC were incubated with a monoclonal antibody to the bovine CD14 molecule 28 followed by goat anti‐mouse immunoglobulin G1 (IgG1)‐coated super‐paramagnetic particles (Miltenyi‐Biotech, Bergisch Gladbach, Germany), and labelled cells were isolated from a MidiMacs column (Miltenyi‐Biotech) according to the manufacturer's instructions. The purity of the cells was evaluated by flow cytometry and shown in each case to be > 98%.…”
