Identification of a Cytopathogenic Toxin from Sneathia amnii

Abstract: Sneathia amnii is a poorly characterized emerging pathogen that has been implicated in amnionitis and urethritis. We found that S. amnii damages fetal membranes, and we identified and purified a cytotoxic exotoxin that lyses human red blood cells and damages cells from fetal membranes. The gene appears to be cotranscribed with a second gene that encodes a protein with identity to two-partner system transporters, suggesting that it is the “A,” or secreted component of a type Vb system. The toxin is 1,881 amino … Show more

“…The species Sneathia amnii and Prevotella amnii have recently been identified as emerging candidates for poor pregnancy outcomes 38 , 39 . In our study, both of these were increased and had a significant association with PTB ( q -value 0.02 and 0.05, respectively; Table 2 ).…”

“…Several of them are known pathogens, like the food-born pathogen Listeria monocytogenes [23]. Other strains are not yet well characterized, but are known to be able to cause infections, for example Sneathia amnii, which can cause urethritis and can affect pregnancy [24]. Whereas Veillonella species belong to the normal flora in the gastrointestinal tract, mouth and vaginal tract and are rarely related to infections [25].…”

“…The role of a cytotoxic haemolysin has recently been unravelled further in 'S. amnii' [57]. The pore-forming cytotoxic exotoxin CptA binds and lyses human red blood cells and damages cells from foetal membranes.…”

“…The secreted component A of the protein toxin of a type Vb system is 1881 aa long with a molecular weight of approximately 200 kDa. It seems to be encoded by a sequence that is cotranscribed with a second gene encoding a protein with identity to two-partner system transporters [57]. The 5642 nt toxin sequence demonstrated in strain ‘ S. amnii ’ Sn35 could also be found in strains DSM 16630 and DSM 16631 T , each with only 84 % homology, but not in S.…”
