Identification and characterization of a putative serine protease expressed in vivo by Mycobacterium avium subsp. paratuberculosis

Abstract: A putative serine protease expressed in vivo by Mycobacterium avium subsp. paratuberculosis was isolated from a lambda g t l l genomic expression library by screening with serum from a naturally infected sheep. The gene was contained in two overlapping clones, which were shown by antibody elution to encode a protein of 34 kDa in M. a. paratuberculosis. The clones were sequenced and database searches detected a motif identical to the active serine site in trypsin, and 30% homology to the putative serine proteas… Show more

“…4. Of these homologs, HtrA (DegP) from E. coli has been purified and shown to have a serine endopeptidase activity (31), in keeping with the presence of the conserved serine protease catalytic domain (the motif GNSGGAL) (3,30,31) which is also present in AlgW. Furthermore, all members of this family of proteins, including AlgW, contain several highly conserved regions, two of which contain invariant His residues and an Asp residue (Fig.…”

“…Since PepA has been shown to be expressed by M. avium subsp. paratuberculosis in infected sheep (14), PepA is likely to be expressed by M. tuberculosis growing in macrophages and to play an important role in facilitating survival. Rv1566c is homologous to the p60 invasion-associated protein of Listeria monocytogenes (37).…”

“…flgal-CA fusion protein was purified by affinity chromatography using a 4 ml column of aminobenzyl 1-thio-fl-galactopyranoside (ABTG) agarose (Sigma) as previously described (Cameron et al, 1994). GST-CA was purified by affinity chromatography using a 4 ml column of glutathione-Sepharose (Pharmacia) as recommended by the manufacturers.…”
